目的:探讨染料木素对阿尔茨海默病模型大鼠学习记忆能力及海马区神经递质含量的影响。方法健康雌性SD大鼠,随机分为5组:假手术组(Sham)、卵巢切除组(Ovx)、卵巢切除+17β-雌二醇组(Ovx+Est)、卵巢切除+17β-雌二醇+染料木素1.0组(Ovx+Est+Gen1.0)、卵巢切除+17β-雌二醇+染料木素10.0组(Ovx+Est+Gen10.0)各6只,给药21 d 后取大鼠脑海马分别测定乙酰胆碱(Ach)、去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)的含量。结果 Ovx组Ach、NE、DA及5-HT递质含量与Sham组比较显著下降(P<0.05);Ovx+Est组Ach、NE、DA及5-HT递质含量与Ovx组比较显著上升( P<0.05);Ovx+Est+Gen1.0组 Ach、NE、DA 及5-HT 递质含量与 Ovx+Est 组相比明显下降( P<0.05), Ovx+Est+Gen10.0组变化与Ovx+Est相似,两组相比无统计学意义。结论雌激素去除降低了脑部主要中枢神经递质的含量,外源性17β-雌二醇可升高中枢神经递质的含量,小剂量染料木素可以拮抗17β-雌二醇的这种作用,高剂量的染料木素对17β-雌二醇的作用无影响。

목적:탐토염료목소대아이자해묵병모형대서학습기억능력급해마구신경체질함량적영향。방법건강자성SD대서,수궤분위5조:가수술조(Sham)、란소절제조(Ovx)、란소절제+17β-자이순조(Ovx+Est)、란소절제+17β-자이순+염료목소1.0조(Ovx+Est+Gen1.0)、란소절제+17β-자이순+염료목소10.0조(Ovx+Est+Gen10.0)각6지,급약21 d 후취대서뇌해마분별측정을선담감(Ach)、거갑신상선소(NE)、다파알(DA)、5-간색알(5-HT)적함량。결과 Ovx조Ach、NE、DA급5-HT체질함량여Sham조비교현저하강(P<0.05);Ovx+Est조Ach、NE、DA급5-HT체질함량여Ovx조비교현저상승( P<0.05);Ovx+Est+Gen1.0조 Ach、NE、DA 급5-HT 체질함량여 Ovx+Est 조상비명현하강( P<0.05), Ovx+Est+Gen10.0조변화여Ovx+Est상사,량조상비무통계학의의。결론자격소거제강저료뇌부주요중추신경체질적함량,외원성17β-자이순가승고중추신경체질적함량,소제량염료목소가이길항17β-자이순적저충작용,고제량적염료목소대17β-자이순적작용무영향。
Objective To study the effects of genistein on learn and memory and the content of neurotransmitter of hippocampus in Alzheimer's disease( AD) rats.Methods Thirty female SD rats were randomized into five groups , each group had six rats: Sham, Ovx, Ovx+Estrogen 1.0 mg/kg body weight (Ovx+Est), Ovx+Estrogen 1.0 mg/kg body weight+Genistein 1.0 mg/kg body weight (Ovx+Est+Gen1.0), Ovx+Estrogen 1.0 mg/kg body weight+Genistein 10.0 mg/kg body weight (Ovx+Est+Gen 10.0), after administration of different estrogens for 21 d, acetylcholine (Ach), norepinephrine(NE), dopamine(DA) and 5-hydroxytryptamine(HT) were measured in hippocam-pus homogenate .Results To compare with those of Sham group , the contents of Ach ,NE,DA and 5-HT in Ovx group were decreased ( P<0.05);To compare with those of Ovx group, the contents of Ach,NE,DA and 5-HT in Ovx+Est group were increased (P<0.05);To com-pare with those of Ovx+Est group, the contents of Ach,NE,DA and 5-HT in Ovx+Est+Gen1.0 groupwere increased (P<0.05); But there was no obvious difference between Ovx+Est+Gen1.0 and Ovx+Est groups.Conclusions Estrogen removal reduces the brain's main central neurotransmitter content, after administration of exogenous 17β-estradiol, the contents of Ach,NE,DA and 5-HT are increased.Low dose of genistein could inhibit this effect of 17β-estradiol.High dose of genistein has no inhibitory effect on 17β-estradiol.
