目的:探讨白细胞介素-6(IL-6 gene ,IL-6)基因启动子区-634C/G多态性与中国广东地区汉族人糖尿病(diabe-tes mellitus,DM)患者视网膜病变(diabetic retinopathy ,DR)的关系。方法在中国广东地区汉族人中,应用聚合酶链反应限制性片段长度多态性(PCR-RFLP )方法,对245例2型糖尿病(T2DM)患者,其中正常眼底即DR0组153例,非增殖期视网膜病变即DR1组51例,增殖期视网膜病变即DR2组41例,和101例健康对照者(NC组)的IL-6基因启动子区-634C/G多态性进行了检测,并比较分析各组间基因型频率和等位基因频率以及相关临床资料。结果(1) DR0、 DR1、 DR2三组间在年龄、性别、血压、体重指数、腰臀比、空腹血糖、餐后2 h血糖、糖化血红蛋白A1 c、血脂等指标均无统计学差异,而病程在三组间差异具有统计学意义(F=19.05,P<0.05)。(2)基因型GG在NC、DR0、DR1、DR2各组间差异有统计学意义(χ2=20.839,P=0.002),DR1、DR2组GG基因型明显高于DR0组,差异有统计学意义(χ2=7.615,P=0.022);等位基因G在NC、DR0、DR1、DR2各组间差异有统计学意义(χ2=11.687,P=0.009),但两组间比较差异无统计学意义。(3)Logistic回归分析表明,IL-6基因启动子区-634G/G基因型、病程及空腹血糖是DR发生的危险因素。结论在中国广东地区汉族人中,IL-6基因启动子区-634 G/G基因型是DR的遗传危险因素。

목적:탐토백세포개소-6(IL-6 gene ,IL-6)기인계동자구-634C/G다태성여중국엄동지구한족인당뇨병(diabe-tes mellitus,DM)환자시망막병변(diabetic retinopathy ,DR)적관계。방법재중국엄동지구한족인중,응용취합매련반응한제성편단장도다태성(PCR-RFLP )방법,대245례2형당뇨병(T2DM)환자,기중정상안저즉DR0조153례,비증식기시망막병변즉DR1조51례,증식기시망막병변즉DR2조41례,화101례건강대조자(NC조)적IL-6기인계동자구-634C/G다태성진행료검측,병비교분석각조간기인형빈솔화등위기인빈솔이급상관림상자료。결과(1) DR0、 DR1、 DR2삼조간재년령、성별、혈압、체중지수、요둔비、공복혈당、찬후2 h혈당、당화혈홍단백A1 c、혈지등지표균무통계학차이,이병정재삼조간차이구유통계학의의(F=19.05,P<0.05)。(2)기인형GG재NC、DR0、DR1、DR2각조간차이유통계학의의(χ2=20.839,P=0.002),DR1、DR2조GG기인형명현고우DR0조,차이유통계학의의(χ2=7.615,P=0.022);등위기인G재NC、DR0、DR1、DR2각조간차이유통계학의의(χ2=11.687,P=0.009),단량조간비교차이무통계학의의。(3)Logistic회귀분석표명,IL-6기인계동자구-634G/G기인형、병정급공복혈당시DR발생적위험인소。결론재중국엄동지구한족인중,IL-6기인계동자구-634 G/G기인형시DR적유전위험인소。
Objective To investigate the association of IL-6 gene promoter region -643C/G polymorphism with diabetic reti-nopathy .Methods The polymorphism of IL-6 gene promoter region -634 C/G in 245 unrelated Guangdong Han people with type 2 di-abetes ,including 153 cases without diabetic retinopathy (DR0 group) ,51 cases without proliferative diabetic retinopathy (NPDR) (DR1 group), 41 cases with proliferative diabetic retinopathy (PDR) (DR2 group), and 101 normal controls(NC group) were detec-ted by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP) methods .The genotype frequency , allele fre-quency and relative clinical data were compared between groups .Results ( 1 ) There were no significant differences in the clinical characteristics except for diabetic duration among DR 0, DR1, DR2 groups.(2) The frequencies of IL-6-643 G/G genotype and G al-lele frequencies among DR0 DR1 DR2 groups were statistically different .There were significantly different in GG genotype between DR0 and DR group,comparison between any two means were not significant differences in G allele frequencies .(3) Logistic regression analysis showed that IL-6 gene promoter region-643G/G genotype,fasting plasma glucose and diabetic duration were risk factors of DR . Conclusions The IL-6 promoter region-643G/G genotype may be a genetic risk factor of DR development .
