目的:探讨芍药苷(paeoniflorin,PAE)对大鼠急性放射性肝损伤(acute radiactive hepatic injury,ARHI)的保护作用。方法按随即区组法将SD雄性大鼠随机分为正常对照组,模型对照组,芍药苷小剂量组(20mg/kg)、中剂量组(40 mg/kg)、大剂量组(80 mg/kg),赤苷组(50 mg/kg)。除正常对照组外,其余各组大鼠均采用VARIN 21-EX直线加速器照射右半肝,其中芍药苷组和赤苷组在照射后每天给予对应剂量药物灌胃,正常对照组、模型对照组给予等容积生理盐水灌胃,各组大鼠在第2、4周末分批取材,取动脉血离心和大鼠右半肝标本10%福尔马林溶液固定备用。分别测定大鼠血清天冬氨酸转氨酶(AST)、谷氨酸转氨酶(ALT),肝组织中谷胱甘肽(GSH)、过氧化物歧化酶(SOD)含量及HE病理染色评分。结果模型对照组于第2周末肝组织HE病理染色评分(2.25±0.53)分升高,AST、ALT分别为(112.83±19.20)U/L、(80.00±21.97)U/L,与正常对照组(63.06±7.15)U/L、(42.30±4.45)U/L比较明显升高(P<0.01)。芍药苷小、中、大剂量组第2周末肝脏GSH含量分别为(60.89±8.43)U/mg、(67.84±9.05)U/mg、(71.92±8.11)U/mg,肝脏SOD含量分别为(118.45±11.98)U/mg、(120.49±13.49)U/mg、(129.65±13.89)U/mg,高于模型对照组(37.32±11.25)U/mg、(90.54±12.12)U/mg,(P<0.05或0.01)。结论受照大鼠在第2周开始出现急性肝损伤,芍药苷对急性放射性肝损伤大鼠有保护作用。

목적:탐토작약감(paeoniflorin,PAE)대대서급성방사성간손상(acute radiactive hepatic injury,ARHI)적보호작용。방법안수즉구조법장SD웅성대서수궤분위정상대조조,모형대조조,작약감소제량조(20mg/kg)、중제량조(40 mg/kg)、대제량조(80 mg/kg),적감조(50 mg/kg)。제정상대조조외,기여각조대서균채용VARIN 21-EX직선가속기조사우반간,기중작약감조화적감조재조사후매천급여대응제량약물관위,정상대조조、모형대조조급여등용적생리염수관위,각조대서재제2、4주말분비취재,취동맥혈리심화대서우반간표본10%복이마림용액고정비용。분별측정대서혈청천동안산전안매(AST)、곡안산전안매(ALT),간조직중곡광감태(GSH)、과양화물기화매(SOD)함량급HE병리염색평분。결과모형대조조우제2주말간조직HE병리염색평분(2.25±0.53)분승고,AST、ALT분별위(112.83±19.20)U/L、(80.00±21.97)U/L,여정상대조조(63.06±7.15)U/L、(42.30±4.45)U/L비교명현승고(P<0.01)。작약감소、중、대제량조제2주말간장GSH함량분별위(60.89±8.43)U/mg、(67.84±9.05)U/mg、(71.92±8.11)U/mg,간장SOD함량분별위(118.45±11.98)U/mg、(120.49±13.49)U/mg、(129.65±13.89)U/mg,고우모형대조조(37.32±11.25)U/mg、(90.54±12.12)U/mg,(P<0.05혹0.01)。결론수조대서재제2주개시출현급성간손상,작약감대급성방사성간손상대서유보호작용。
Objective Investigate the Protective effect of paeoniflorin in rats with acute liver injury. Methods Male SD rats were randomly divided into normal group, model group, paeoniflorin in small, middle and high dose group (20 mg/kg, 40 mg/kg, 80 mg/kg), paeony glycoside group (50 mg/kg). Except normal group, the rest of groups were irradiated fractionally by VARIN 21-EX linear accelerator at right liver, The paeoniflorin group and paeony glycoside group were lavaged everyday after irradiation for corresponding drugs and doses. Normal group and model group give equal volume normal saline everyday. All rats were killed on 2nd and 4th weekend. Measure rats serum AST, ALT, hepatic tissue GSH, SOD, and HE staining score. Results The rats in model group liver tissue HE staining scores increased to(2.25±0.53)on 2nd weekend, The serum levels of AST, ALT increased to(112.83±19.20)U/L, (80±21.97)U/L, it significantly increased Compared with the normal group(63.06±7.15)U/L, (42.30±4.45)U/L, P<0.01. The liver GSH contents of paeoniflorin in each dosage groups rats were(60.89±8.43)U/mg, (67.84±9.05)U/mg, (71.92±8.11)U/mg on 2 nd weekend, Compared with the model group(37.32±11.25)U/mg, (90.54±12.12)U/mg, P<0.05或<0.01. Conclusions The irradiated rats go into acute liver injury on 2nd weekend, paeoniflorin has protective effect on acute liver injury in rats.
