目的：探讨安徽汉族超重者与正常体重者脂肪组织中 TNF-α基因的差异表达，分析 TNF-α基因的表达与年龄（AGE）、体重指数(BMI)、腰围尺寸(WC)、腰臀比例(WHR)的相关性。方法以肌动蛋白（β-actin）基因作为内参照，用半定量RT-PCR方法检测网膜脂肪组织与皮下脂肪组织的TNF-α基因在24例正常体重者（BMI在18.5～24 kg/m2之间）和19例超重者(BMI 在24～28 kg/m2之间)中的不同表达水平，并通过测量实验者的一系列指标，有体重、身高、臀围、腰围等，在此基础上计算出腰臀比例和体重指数，分析比较这些参数与TNF-α基因表达的相关性。结果腹部皮下与网膜脂肪组织中的TNF-α基因表达水平在超重组与正常体重组间差异均无显著性；超重组组内与正常体重组组内的皮下及网膜脂肪的TNF-α基因表达差异亦无显著性；WHR、WC与皮下及网膜的TNF-α基因表达相关性为正向。结论皮下及网膜脂肪组织TNF-α基因表达在超重组中无明显增强，但与WHR、WC呈正相关。
목적：탐토안휘한족초중자여정상체중자지방조직중 TNF-α기인적차이표체，분석 TNF-α기인적표체여년령（AGE）、체중지수(BMI)、요위척촌(WC)、요둔비례(WHR)적상관성。방법이기동단백（β-actin）기인작위내삼조，용반정량RT-PCR방법검측망막지방조직여피하지방조직적TNF-α기인재24례정상체중자（BMI재18.5～24 kg/m2지간）화19례초중자(BMI 재24～28 kg/m2지간)중적불동표체수평，병통과측량실험자적일계렬지표，유체중、신고、둔위、요위등，재차기출상계산출요둔비례화체중지수，분석비교저사삼수여TNF-α기인표체적상관성。결과복부피하여망막지방조직중적TNF-α기인표체수평재초중조여정상체중조간차이균무현저성；초중조조내여정상체중조조내적피하급망막지방적TNF-α기인표체차이역무현저성；WHR、WC여피하급망막적TNF-α기인표체상관성위정향。결론피하급망막지방조직TNF-α기인표체재초중조중무명현증강，단여WHR、WC정정상관。
Objective To investigate the expression of tumor necrosis factor alpha in adipose tissues of Anhui Han nationality be-tween over-weight and control group, and the relationship with AGE, BMI, WC, WHR. Methods The TNF-α expression level was measured by semi-quantitative RT-PCR in abdominal subcutaneous and omental adipose tissues of 24 normal controls (BMI:18.5~24 kg/m2) and 19 over-weights (BMI:24~28 kg/m2). Anthropometric data, such as height, body weight, waist circumference, hip circumference;and waist to calculate waist-hip ratio (WHR) and body mass index (BMI). Insulin resistance index were calcu-lated. Compare these parameters with TNF- α gene expression by correlation analysis. Results The level of TNF-α gene expres-sion have no significant difference between overweight and control group, respectively;There were no differences between subcuta-neous and omental adipose tissues from each subject. WHR and WC were found to have significant positive correlation with the expression of TNF-α gene in subcutaneous and omental adipose tissues. Conclusion The gene expression levels of TNF-α was positively correlated with WHR and WC, but have no obviously increasing in the over-weight group when compared with the con-trol group.
