对分离的血红蛋白抗菌肽进行生物活性鉴定。通过微量稀释法测定血红蛋白抗菌肽对大肠埃希菌、金黄色葡萄球菌、白色念珠菌的最小抑菌浓度(MIC);以血红蛋白释放量测定其溶血性;琼脂平皿试验测定其热稳定性;并进一步在扫描电镜(SEM)下观察血红蛋白抗菌肽对测试菌株形态结构的影响。结果表明,血红蛋白抗菌肽对大肠埃希菌、金黄色葡萄球菌和白色念珠菌的 MIC 分别为0.5、1.0、0.25 mg/mL;在2.0 mg/mL 的浓度下,可造成6.7%的红细胞发生溶血;经过高温处理,仍表现出较好的热稳定性;SEM 观察到血红蛋白抗菌肽作用的细菌形态发生严重的皱缩、变形并大量的聚集,细胞膜裂解、破碎。说明血红蛋白抗菌肽具有良好的抗菌活性且溶血性较低,具有良好的生物安全性。

To analyze the bioactivity of novel hemoglobin antimicrobial peptide,the minimal inhibitory con-centration of hemoglobin antimicrobial peptide to Escherichia coli ,Staphylococcus aureus,Monilia albican was tested by broth microdilution method.The hemolysis was tested by measuring the release of hemoglo-bin.Thermal stability of hemoglobin antimicrobial peptide was determined by agar gel diffusion method. And the morphology of bacteria tesed were observed by scanning electron microscope.The results showed that the MIC to Escherichia coli ,Staphylococcus aureus ,Monilia albican were 0.5,1.0,0.25 mg/mL. It caused 6.7% rupture of erythrocyte at the concentration of 2.0 mg/mL.After heated,it kept stability partly.The shrinking,distorting and gathering of microorganism were observed.It indicated that hemo-globin antimicrobial peptide had a good bio-safety in the development and application,which had a signifi-cant antibacterial activity and low hemolysis.
