为调查潍坊地区犬细小病毒的生物学特性和基因变异情况,收集了该地区2005年-2013年16份不同来源的犬细小病毒病料,通过 F81细胞分离、PCR 扩增、电镜观察、血凝试验、人工感染、VP2基因克隆测序等方法进行了生物学特性和分子流行病学调查。结果显示,分离到的5个地方毒株的生物学特性并未改变:能够凝集猪红细胞,对其他动物红细胞不凝集;5个毒株均能导致试验犬感染发病;2个毒株的基因型为 CPV-2a,3个毒株为 CPV-2b,氨基酸同源性和进化树分析表明,5个地方毒株与国际参考毒株的同源性在96%以上,与国内北京分离株亲缘关系最近,说明该地区的毒株进化与世界范围内其他毒株保持了同步和一致。

To research the biological characteristics and gene variation of canine parvovirus (CPV)in Wei-fang surroundings,16 specimens from different dogs infected with CPV were collected during 2005 to 2013 and they were studied by series of experiments such as viral isolation in F81,PCR,electronic microscope, HA/HI test,experimental infection and gene sequencing of vp2.Five local CPV strains were isolated from these specimens,in which two strains were proved to be CPV-2a and other three strains were CPV-2b. These 5 virus strains could agglutinate 1% swine erythrocyte.The experimental dogs showed typical clini-cal symptoms by artificial infection with five local strains.Conpared with other international reference strains,the amino acid homology of these five local strains were higher than 96% and shared the highest homology with Beijing isolate(CPV-02/09).Results showed that CPV in Weifang areas keeped the same mutation pace and mode with other strains in worldwide.
