目的:探讨输入人脐带间充质干细胞(human umbilical cord mesenchymal stem cells, HUMSCs)对脂多糖(LPS)诱导的大鼠 ALI 的治疗作用。方法将30只 Wistar 大鼠随机分为3组:正常对照组、ALI 模型组和 HUMSCs 移植组,每组10只。健康大鼠 LPS 气管滴入方法建立 ALI 模型,尾静脉输入 HUMSCs。输入7 h 后,处死各组大鼠,取肺组织行病理学观察,测定肺湿/干重比(W/D), ELISA 法测定肺组织匀浆中肿瘤坏死因子α(TNF-α)和 IL-1β的含量。结果肺组织病理学显示:ALI模型组大鼠肺组织呈现典型的炎症病理变化,包括肺泡充血、水肿、出血,肺泡腔和血管壁中性粒细胞浸润,肺泡壁增厚等肺损伤性病变;HUMSCs 移植组大鼠肺组织损伤程度明显减轻。与正常对照组比较, ALI 模型组 W/D、TNF-α和 IL-1β含量明显升高(P <0.05)。与 ALI 模型组比较,HUMSCs 移植组 W/D、TNF-α和 IL-1β含量明显降低(P <0.05)。结论输入 HUMSCs 能减轻 ALI,并能降低肺 W/D 和肺组织 TNF-α及 IL-1β含量。

목적:탐토수입인제대간충질간세포(human umbilical cord mesenchymal stem cells, HUMSCs)대지다당(LPS)유도적대서 ALI 적치료작용。방법장30지 Wistar 대서수궤분위3조:정상대조조、ALI 모형조화 HUMSCs 이식조,매조10지。건강대서 LPS 기관적입방법건립 ALI 모형,미정맥수입 HUMSCs。수입7 h 후,처사각조대서,취폐조직행병이학관찰,측정폐습/간중비(W/D), ELISA 법측정폐조직균장중종류배사인자α(TNF-α)화 IL-1β적함량。결과폐조직병이학현시:ALI모형조대서폐조직정현전형적염증병리변화,포괄폐포충혈、수종、출혈,폐포강화혈관벽중성립세포침윤,폐포벽증후등폐손상성병변;HUMSCs 이식조대서폐조직손상정도명현감경。여정상대조조비교, ALI 모형조 W/D、TNF-α화 IL-1β함량명현승고(P <0.05)。여 ALI 모형조비교,HUMSCs 이식조 W/D、TNF-α화 IL-1β함량명현강저(P <0.05)。결론수입 HUMSCs 능감경 ALI,병능강저폐 W/D 화폐조직 TNF-α급 IL-1β함량。
Objective To investigate the therapeutic effect of human umbilical cord mesenchymal stem cells (HUMSCs ) on acute lung injury (ALI ) induced by lipopolysaccharide (LPS) in rats. Methods HUMSCs were isolated,cultured,and identified.30 Wistar rats were randomly divided into three groups (n = 10 ):normal control group (group A ),ALI group (group B ),and HUMSCs transplantation group (group C).Healthy rats were intratracheally given LPS to establish ALI models, and the HUMSCs were given to rats via caudal vein.After 7 h of transplantation,rats were sacrificed and lung tissues were collected and stored according to the protocol.Pathological changes of lung tissue were observed under light microscope.Besides,ratio of lung wet and dry weight (W/D)and levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in lung tissue homogenate were measured.Results Histopathological evaluation showed that alveolar congestion,edema,hemorrhage,infiltration of neutrophils in the airspace or vessel wall and thickness of the alveolar wall,were observed in group B. However,these changes were obviously relieved in group C.Compared with group A,lung W/D and levels of TNF-αand IL-1βwere significantly increased in group B (P <0.05).In group C,lung W/D and levels of TNF-α and IL-1β were significantly decreased compared to group B (P < 0.05 ).Conclusions Transplantation of HUMSCs can attenuate LPS-induced ALI and decrease the levels of TNF-αand IL-1βin rats.
