薄层色谱分离效果的好坏,很大程度上取决于展层剂的组成和配比,还会受硅胶板的活化度、环境温度、显色剂和点样量手法等因素的影响。本文报道了采用薄层色谱法分析玉米芯降解液中单糖的成分,着重对多种展层剂进行优化研究。研究结果显示以正丁醇∶乙酸乙酯∶水=7∶2∶1为展层剂分离糖的效果较好,本文还显示以苯胺-二苯胺-磷酸为显色剂的糖显色效果较好。同时,用上述优化的方法对不同酸浓度处理的玉米芯降解液进行分析研究,可以准确、清晰、便利地定性检测出玉米芯木聚糖中单糖的组分。
薄層色譜分離效果的好壞,很大程度上取決于展層劑的組成和配比,還會受硅膠闆的活化度、環境溫度、顯色劑和點樣量手法等因素的影響。本文報道瞭採用薄層色譜法分析玉米芯降解液中單糖的成分,著重對多種展層劑進行優化研究。研究結果顯示以正丁醇∶乙痠乙酯∶水=7∶2∶1為展層劑分離糖的效果較好,本文還顯示以苯胺-二苯胺-燐痠為顯色劑的糖顯色效果較好。同時,用上述優化的方法對不同痠濃度處理的玉米芯降解液進行分析研究,可以準確、清晰、便利地定性檢測齣玉米芯木聚糖中單糖的組分。

Effect of thin -layer chromatography mainly depends on the compose and proportion of developing solvents , as well influenced by silica gel plate activation grade ,environment temperature , color developing agentand and spotting technique .This paper reports that thin -layer chromatography analyze monosaccharide ingredients of the corncob degradation solution , emphasizing optimization study on developing solvent of the method .The result shows that thin -layer chromatography can be effective to separate the saccharides by Vn-butanol∶Vethyl acetate∶VWater =7∶2∶1 as developing solvent .The result also shows saccharides has better color effect by aniline -diphenylamine-phosphoric acid as chromogenic reagent .Meanwhile , The method of the optimization above analyzes degradation solution of different acid concentrations treatment corncob , which can effectively determination of the type of saccharides.Thus, the method can be accurate, clear, convenient qualitative detection of corncob xylan monosaccharide components .
