目的:研究人卵巢颗粒细胞胰岛素受体(insulin receptor,INSR)和胰岛素样生长因子-1受体(insulin-like growth factor 1 receptor,IGF-1R)的表达及相关性。方法接受体外受精-胚胎移植( in vitro fertilization-embryo transfer,IVF-ET)不孕症患者84例,按照妊娠结果分为妊娠组和未妊娠组,每组42例。应用实时荧光定量PCR( real-time PCR)法测定取排卵日成熟卵泡壁颗粒细胞INSR,IGF-1R mRNA的表达;并对INSR,IGF-1R mRNA之间及与血清LH、E2、P水平相关性分析。结果妊娠组血清雌二醇水平高于未妊娠组( P <0照.05)。妊娠组优质胚胎数高于未妊娠组( P <0.05)。其余两组比较,差异无统计学意义( P >0.05)。妊娠组INSR、IGF-1R、表达高于未妊娠组( P <0.05)。INSR与IGF-1R呈正相关( r =0.712, P <0.01)。结论颗粒细胞INSR、IGF-1R可能参与卵母细胞质量的调节。

Objective To investigate the expression of insulin receptor in human ovary granulosa cells ( INSR) and insulin-like growth factor 1 receptor (IGF-1R) and their correlation. Methods Eighty-four patients with infertility received in vitro fertilization and embryo transplantation ( IVF-ET ) , the patients were divided into pregnancy groups ( n =42 ) and non-pregnancy group ( n =42).The expressions levels of INSR and IGF-1R mRNA in mature follicle wall granulosa cells were detected by Real-time PCR.The correlation between INSR , IGF-1R and serum sex hormones was analyzed .Results The high-quality embryo numbers in pregnancy groups were significantly higher than those in non -pregnancy group ( P <0 .05 ) . However there were no significant differences in the other sex hormones levels including LH ,E2,P between the two groups ( P>0.05).The levels of INSR and IGF-1R in pregnancy groups were significantly higher than those in non-pregnancy group ( P<0.05).Moreover the expression of INSR was positively correlated to that of IGF-1R ( r =0.712, P <0.01).Conclusion INSR in human ovary granulosa cells and IGF-1R may be involved in the regulation of oocyte quality .
