目的:探讨模拟失重后再超重环境对猴咬肌细胞RyR1 mRNA表达的影响。方法:健康雄性猕猴23只,随机分为7组:正常对照组(A组)、模拟失重(B组)、模拟超重(C组)以及失重后再超重+11 Gx/270 s(D1组)、+13Gx/230 s(D2组)、+15Gx/200 s(D3组)(以上各组恢复2 d)、+13Gx/230 s组(D4组)(恢复10 d);各组进行相应模拟实验后处死动物,取其咬肌组织制作冰冻切片,HE染色法观察咬肌细胞的组织形态学变化;RT-PCR法检测咬肌细胞中RyR1 mRNA的表达水平。结果:①单纯失重和单纯超重组咬肌细胞胞膜边界模糊不清,横纹不清晰或消失,间质稀疏;模拟失重后再超重各组部分肌细胞肿胀,偶见细胞核呈扁平状;②各实验组(除D4组外)咬肌细胞中的RyR1 mRNA表达水平均明显下降(P<0.05);失重后再超重各组分别与单纯超重组(C组)相比,其中D2、D3组的RyR1 mRNA表达水平明显降低(P<0.05);而D4组的RyR1 mR-NA表达水平则有所上升,并高于D2组(P<0.05)。结论:一定范围失重、超重和失重后再超重环境均可不同程度地造成咬肌细胞组织形态学改变,并可引起RyR1 mRNA表达水平的下降,这些改变是可恢复的。

목적:탐토모의실중후재초중배경대후교기세포RyR1 mRNA표체적영향。방법:건강웅성미후23지,수궤분위7조:정상대조조(A조)、모의실중(B조)、모의초중(C조)이급실중후재초중+11 Gx/270 s(D1조)、+13Gx/230 s(D2조)、+15Gx/200 s(D3조)(이상각조회복2 d)、+13Gx/230 s조(D4조)(회복10 d);각조진행상응모의실험후처사동물,취기교기조직제작빙동절편,HE염색법관찰교기세포적조직형태학변화;RT-PCR법검측교기세포중RyR1 mRNA적표체수평。결과:①단순실중화단순초중조교기세포포막변계모호불청,횡문불청석혹소실,간질희소;모의실중후재초중각조부분기세포종창,우견세포핵정편평상;②각실험조(제D4조외)교기세포중적RyR1 mRNA표체수평균명현하강(P<0.05);실중후재초중각조분별여단순초중조(C조)상비,기중D2、D3조적RyR1 mRNA표체수평명현강저(P<0.05);이D4조적RyR1 mR-NA표체수평칙유소상승,병고우D2조(P<0.05)。결론:일정범위실중、초중화실중후재초중배경균가불동정도지조성교기세포조직형태학개변,병가인기RyR1 mRNA표체수평적하강,저사개변시가회복적。
AIM:To study the effects of high +Gx exposure after simulated weightlessness on RyR1 mRNA expression in masseter muscle of Rhesus macaque.METHODS:23 male Rhesus macaques were randomly divided in-to 4 groups:control (A),simulated weightlessness (B),simulated overweight (C)and high +Gx exposure after sim-ulated weightlessness (D)groups.Group D was further divided into 4 subgroups according to the Gx loads and dura-tion,including +11Gx/270s (D1),+13Gx/230s (D2),+15Gx/200s (D3)2 and 10 day recovery after exposure in +13Gx/230s (D4)groups.Masseter muscle was sampled and obsserved by HE staining and RT-PCR.RE-SULTS:Histopathological observation showed that the sarcolemma of the experimental groups exhibited fuzzy bounda-ries.The striation were not clear or disappeared.Interstitial space was slightly sparse and some muscle cells were swol-len.Occasionally nucleus were flat or shifted in the center.RyR1 mRNA expression in masseter muscle of group B, group C,group D1,D2 and D3 was significantly decreased (P<0.05).RyR1 mRNA expression in D2 and D3 groups was significantly lower than in control group (P<0.05).However,RyR1 mRNA in D4 group was significantly higher than that in D2 group (P<0.05 ).CONCLUSION:Simulated weightlessness,simulated overweight and high +Gx exposure after simulated weightlessnes tested in this study can cause pathological changes and down-regulate RyR1 mR-NA expression in masseter muscle cells of Rhesus macaque.The changes are reversibe.
