目的：观察雷公藤内酯醇（ Triptolide，TPL）和奥沙利铂（ Oxaliplatin，L-OHP）单用及联合应用对人结肠癌细胞 SW480体外增殖的影响，初步探讨其作用机理。方法 MTT法检测L-OHP和TPL单用和联合对人结肠癌细胞株SW480增殖的抑制作用；流式细胞仪检测细胞周期改变。结果 TPL和L-OHP联合应用对SW480细胞的增殖有协同抑制作用。流式细胞仪检测发现细胞阻滞于S期，G2/M期细胞减少及出现凋亡峰；结论实验证实TPL和L-OHP联合应用能协同抑制SW480细胞增殖，其作用机理有可能是通过阻滞细胞周期引起的。
목적：관찰뢰공등내지순（ Triptolide，TPL）화오사리박（ Oxaliplatin，L-OHP）단용급연합응용대인결장암세포 SW480체외증식적영향，초보탐토기작용궤리。방법 MTT법검측L-OHP화TPL단용화연합대인결장암세포주SW480증식적억제작용；류식세포의검측세포주기개변。결과 TPL화L-OHP연합응용대SW480세포적증식유협동억제작용。류식세포의검측발현세포조체우S기，G2/M기세포감소급출현조망봉；결론실험증실TPL화L-OHP연합응용능협동억제SW480세포증식，기작용궤리유가능시통과조체세포주기인기적。
OBJECTIVE To observe the growth inhibitory effect of Oxaliplatin ( L-OHP) and Triptolide ( TPL) on proliferation in human colon carcinoma cell lines SW 480 in vitro and investigate the mechanism.METHODS The growth inhibitory effect was evaluated by MTT assay in single agent or combination treatment.Distribution of cell cycle and apoptosis was analyzed by using flow cytometry.RESULTS Combination treatment of TPL and L-OHP has synergistic effect on killing SW 480 cells.The analysis of cell cycle indicated that TPL and L-OHP blocked cells at S phases and cells of G 2/M phase reduced.CONCLUSION Combination treatment of TPL and L-OHP could in-hibit the proliferation of SW480 cells obviously according to the experiment ,which mechanism probably is a synergis-tic interaction in the cell cycle.
