目的:探讨中药麝香保心丸对大鼠放射性肺损伤(RILI)的防治作用。方法:将100只清洁级SD大鼠完全随机分为3组:对照组、单纯照射组、治疗组(照射+麝香保心丸)。应用SD大鼠建立放射性肺损伤模型,治疗组自照射前3天至照射后15天给予麝香保心丸灌胃,于照射后第3天、7天、15天、30天、60天分别处死每组大鼠6只,取心脏血,ELSA法检测血浆内皮素(endothelin-1,ET-1)含量变化,取肺组织通过HE染色观察炎性渗出,免疫组化检测Ⅰ型胶原纤维生成情况。结果:照射后各组大鼠肺组织炎性渗出、纤维化逐渐加重,单纯照射组最严重。照射后第3天、7天、15天Ⅰ型胶原纤维表达量各组间无明显差异(P>0.05),30天时明显增加,60天时稍减少,治疗组较单纯照射组表达减弱(P<0.05)。血浆ET-1含量自照射后3天逐渐增加,至30天时达高峰,60天时稍下降,治疗组比单纯照射组含量明显减少(P<0.01)。结论:麝香保心丸可以减轻RILI大鼠肺组织炎性渗出、抑制胶原纤维合成,对RILI有一定的防治作用。

목적:탐토중약사향보심환대대서방사성폐손상(RILI)적방치작용。방법:장100지청길급SD대서완전수궤분위3조:대조조、단순조사조、치료조(조사+사향보심환)。응용SD대서건립방사성폐손상모형,치료조자조사전3천지조사후15천급여사향보심환관위,우조사후제3천、7천、15천、30천、60천분별처사매조대서6지,취심장혈,ELSA법검측혈장내피소(endothelin-1,ET-1)함량변화,취폐조직통과HE염색관찰염성삼출,면역조화검측Ⅰ형효원섬유생성정황。결과:조사후각조대서폐조직염성삼출、섬유화축점가중,단순조사조최엄중。조사후제3천、7천、15천Ⅰ형효원섬유표체량각조간무명현차이(P>0.05),30천시명현증가,60천시초감소,치료조교단순조사조표체감약(P<0.05)。혈장ET-1함량자조사후3천축점증가,지30천시체고봉,60천시초하강,치료조비단순조사조함량명현감소(P<0.01)。결론:사향보심환가이감경RILI대서폐조직염성삼출、억제효원섬유합성,대RILI유일정적방치작용。
Objective:To investigate the protection of Shexiang Baoxin Pill on radiation-induced lung injury(ra-diation induced lung injury,RILI).Methods:All 100 SD rats were randomly divided into 3 groups:control group,ra-diation alone group,treatment group(irradiated and treated with Shexiang Baoxin Pill).These rats were radiated to make the modle of RILI.Since 3 days before radiation to 15 days after radiation,the treatment group was gavaged with Shexiang Baoxin Pill.Then 6 rats of each group were put to death in the days 3rd,7th,15th,30th,60th after radiation. Frist,blood serum was collected from hearts to check endothelin-1 (ET-1 )by ELISA;Secondly,changes of inflam-mation and type Ⅰ collagen fiber were observed by HE staining and immunohistochemical method.Results:Pneumo-nia and fibrosis in the rats which were radiated were worse by worse,especially in the radiation alone group.The level ofⅠCollagen fiber was no distinctly difference in three groups after radiation 3rd,7th,15th(P>0.05),then 30th day it was increased to max,60th day was decreased.And at these two time points treatment group was abviously lower than radiation alone group(P<0.05).The ET-1 content was added step by step after radiation,30th day after radiation to max,while 60th day degreased.The quantity of ET-1 in the treatment group was increased less than radiation alone group(P<0.01).Conclusion:Shexiang Baoxin Pill may relieve inflammation and fibrosis in the RILI of rats to pre-vent and treat RILI.
