目的:探讨蜕皮甾酮对2型糖尿病大鼠肝细胞胰岛素受体底物2( IRS-2)蛋白表达的影响。方法选择42只大鼠,14只作为正常组,常规饲料喂养,余下28只先以高脂喂养加小剂量STZ注射诱导2型糖尿病大鼠模型,造模成功后,随机分为模型组和治疗组,分别给予高脂饲料喂养、高脂饲料喂养+蜕皮甾酮灌胃治疗。5周后取大鼠肝组织标本,应用免疫组化和RT-PCR技术检测IRS-2蛋白和mRNA含量。结果正常组和治疗组肝细胞IRS-2蛋白及mRNA含量均明显高于模型组( P<0.05或<0.01)。结论蜕皮甾酮可提高2型糖尿病大鼠肝细胞IRS-2蛋白和mRNA含量,这可能是蜕皮甾酮改善2型糖尿病大鼠胰岛素抵抗的机制之一。

목적:탐토세피치동대2형당뇨병대서간세포이도소수체저물2( IRS-2)단백표체적영향。방법선택42지대서,14지작위정상조,상규사료위양,여하28지선이고지위양가소제량STZ주사유도2형당뇨병대서모형,조모성공후,수궤분위모형조화치료조,분별급여고지사료위양、고지사료위양+세피치동관위치료。5주후취대서간조직표본,응용면역조화화RT-PCR기술검측IRS-2단백화mRNA함량。결과정상조화치료조간세포IRS-2단백급mRNA함량균명현고우모형조( P<0.05혹<0.01)。결론세피치동가제고2형당뇨병대서간세포IRS-2단백화mRNA함량,저가능시세피치동개선2형당뇨병대서이도소저항적궤제지일。
Objective To investigate the effect of ecdysterone on the expression of insulin receptor substrate 2 ( IRS-2 ) in liver cell of type 2 diabetic rats .Methods 14 of the 42 rats were randomly selected to the normal control group , and was given ordinary rat food .The other 28 rats were made into type 2 diabetes mellitus rat models by using the high fatty diet and small dose of SIZ , and then diveided into model group and treatment group .The rats in model group or treatment group was given high fatty diet or high fatty diet fecdysterone .IRS-2 protein and mRNA in liver tissues of rats were detected by immunohistochemistry assay and RT-PCR assay after five weeks .Results The expression of IRS-2 protein and mRNA in normal group and treatment group were higher than those of the model group (P<0.01or <0.01).Conclusions Ecdyste-rone led to up-regulation expression of IRS-2 protein and IRS-2 mRNA of liver tissue in diabetic rats .This might play a role in ecdysterone-improved hepatic insulin resistance .
