目的:观察五味子多糖对人脑胶质瘤细胞生长的抑制作用,初步探讨五味子多糖作用肿瘤细胞的机制。方法通过体外细胞实验的方式,培养人脑胶质瘤SHG-44细胞,将其分为空白组和用药组,倒置显微镜观察五味子多糖作用后的凋亡情况,用WST-1法检测人脑胶质瘤细胞中SOD和GSH的含量,ELISA法检查凋亡蛋白Bcl-2、bax和Caspase-3的表达情况。结果通过与用药组进行比较,倒置显微镜下观察不同浓度的五味子多糖作用于脑胶质瘤SHG-44细胞48 h可明显抑制其生长;与对照组相比,肿瘤细胞中SOD和GSH含量变化具有统计学意义(P<0.01),含量轻度降低;Caspase-3和Bcl-2/bax含量明显降低。结论五味子多糖可以在一定程度上促进脑胶质瘤细胞凋亡。

목적:관찰오미자다당대인뇌효질류세포생장적억제작용,초보탐토오미자다당작용종류세포적궤제。방법통과체외세포실험적방식,배양인뇌효질류SHG-44세포,장기분위공백조화용약조,도치현미경관찰오미자다당작용후적조망정황,용WST-1법검측인뇌효질류세포중SOD화GSH적함량,ELISA법검사조망단백Bcl-2、bax화Caspase-3적표체정황。결과통과여용약조진행비교,도치현미경하관찰불동농도적오미자다당작용우뇌효질류SHG-44세포48 h가명현억제기생장;여대조조상비,종류세포중SOD화GSH함량변화구유통계학의의(P<0.01),함량경도강저;Caspase-3화Bcl-2/bax함량명현강저。결론오미자다당가이재일정정도상촉진뇌효질류세포조망。
Objective To observe the inhibitory effect of Polysaccharide from Schisandra chinensis on the growth of human glioma cells ,and study its mechanism of antitumor .Methods SHG-44 glioma cells were divided into control and treated groups .The morphology of apoptosis were observed by inverted microscope .WST-1 assay was used to eval-uate SOD and GSH.The secretion of Bcl-2,bax and caspase 3 were detected by ELISA.Results The different con-centrations of FSP could significantly inhibit the growth of glioma cells SHG-44 at 48 h.Compared with the control group,SOD and GSH content in the treated group was lightly lower ,and the expression of Caspase-3 and Bcl-2/bax al-so appeared the much lower trend ,which had statistical significance .Conclusion FSP can promote the apoptosis of glioma cells to a certain extent .
