通过对提取溶剂、流动相条件、最大吸收波长及色谱条件的优化,建立了一种同时测定葡萄酒中坚牢红、偶氮玉红、孟加拉红、奇通红S、荧光桃红5种酸性红色素的高效液相色谱分析方法。样品经甲醇提取后,采用Symmetry C18柱进行分离,以甲醇-20 mmol/L醋酸铵为流动相梯度洗脱,二极管阵列检测器检测,外标法定量。结果表明,各组分在0.5~50.0 mg/L范围内线性关系良好,相关系数均大于0.999,检出限为0.1~1.0 mg/kg;在1、10、50 mg/kg三个加标水平下的平均回收率为85%~106%,相对标准偏差( RSD)为1.5%~5.8%。

A high performance liquid chromatographic ( HPLC) method was developed for simultaneously detecting 5 acid red pigments such as fast red , azorubine , rose bengal , kiton red S and phloxine B in grape wine .The extracting solvent , mobile phase type , maximum absorption wavelength and chromatographic condi-tions were optimized through experiment .The sample was extracted with methanol , and the extract was separa-ted through a Symmetry C18 column (25 cm ×4.6 mm, 5μm) using methanol-20 mmol/L ammonium ace-tate as mobile phase by gradient elution .The target compounds were confirmed by diode -array detection and quantified by external standard method .The calibration curves of 5 pigments showed good linearity in the range of 0.5~50.0 mg/L with the correlative coefficients more than 0.999.The detection limits were in the range of 0.1~1.0 mg/kg.The mean recovery at 3 spiked levels of 1, 10, 50 mg/kg ranged from 85% to 106%with the RSD as 1 .5%~5 .8%.
