目的:观察中药红景天提取物对高脂饮食诱导的胰岛素抵抗大鼠胰岛素敏感性及过氧化物酶体增殖物激活受体-γ( PPAR-γ) mRNA表达的影响。方法将70只Wistar大鼠随机分为2组,正常对照组20只,高脂模型组50只。正常对照组给予基础饲料,高脂模型组给予高脂饲料,均喂养4周后2组各随机选取10只大鼠行高胰岛素-正葡萄糖钳夹试验判断造模是否成功,确认造模成功后将剩余高脂模型组大鼠随机分为高脂对照组和高脂+红景天干预组,每组20只。高脂+红景天干预组给予红景天提取物1 g/( kg · d)灌胃,正常对照组及高脂对照组给予等容积0.9%氯化钠注射液灌胃,其他饲养条件不变,继续喂养4周。观察比较各组大鼠造模后和灌胃4周后血清甘油三酯( TG)、血清总胆固醇( TC)、游离脂肪酸( FFA)、空腹血糖( FPG)及空腹血清胰岛素( FINS)的含量变化,并记录葡萄糖输注率( GIR)变化及PPAR-γmRNA的表达情况。结果高脂模型组大鼠造模后与正常对照组比较FPG、FINS明显升高(P<0.05),GIR明显下降(P<0.05);高脂对照组灌胃4周后与正常对照组比较TG、TC及FFA水平均明显升高(P<0.05),高脂+红景天干预组较高脂对照组TC、TG及FFA水平均明显下降(P<0.05),与正常对照组水平相当(P>0.05);高脂对照组灌胃4周后与正常对照组比较FPG、FINS明显升高(P<0.05),GIR明显下降(P<0.05),高脂+红景天干预组较高脂对照组FPG、FINS水平明显下降(P<0.05),GIR明显升高(P<0.05),与正常对照组水平相当相(P>0.05);高脂对照组灌胃4周后PPAR-γmRNA表达水平与正常对照组表达水平相当(P>0.05),高脂+红景天干预组较正常对照组及高脂对照组比较PPAR-γmRNA表达水平均有明显升高(P<0.05)。结论红景天提取物对大鼠的胰岛素抵抗有显著地改善作用,降低血清TG、TC及FFA明显降低,调节FPG平衡,降低FINS水平,增加GIR,提高胰岛素敏感性,提示其作用机制可能与提高胰岛素抵抗大鼠肝脏细胞PPAR-γmRNA表达有关。
目的:觀察中藥紅景天提取物對高脂飲食誘導的胰島素牴抗大鼠胰島素敏感性及過氧化物酶體增殖物激活受體-γ( PPAR-γ) mRNA錶達的影響。方法將70隻Wistar大鼠隨機分為2組,正常對照組20隻,高脂模型組50隻。正常對照組給予基礎飼料,高脂模型組給予高脂飼料,均餵養4週後2組各隨機選取10隻大鼠行高胰島素-正葡萄糖鉗夾試驗判斷造模是否成功,確認造模成功後將剩餘高脂模型組大鼠隨機分為高脂對照組和高脂+紅景天榦預組,每組20隻。高脂+紅景天榦預組給予紅景天提取物1 g/( kg · d)灌胃,正常對照組及高脂對照組給予等容積0.9%氯化鈉註射液灌胃,其他飼養條件不變,繼續餵養4週。觀察比較各組大鼠造模後和灌胃4週後血清甘油三酯( TG)、血清總膽固醇( TC)、遊離脂肪痠( FFA)、空腹血糖( FPG)及空腹血清胰島素( FINS)的含量變化,併記錄葡萄糖輸註率( GIR)變化及PPAR-γmRNA的錶達情況。結果高脂模型組大鼠造模後與正常對照組比較FPG、FINS明顯升高(P<0.05),GIR明顯下降(P<0.05);高脂對照組灌胃4週後與正常對照組比較TG、TC及FFA水平均明顯升高(P<0.05),高脂+紅景天榦預組較高脂對照組TC、TG及FFA水平均明顯下降(P<0.05),與正常對照組水平相噹(P>0.05);高脂對照組灌胃4週後與正常對照組比較FPG、FINS明顯升高(P<0.05),GIR明顯下降(P<0.05),高脂+紅景天榦預組較高脂對照組FPG、FINS水平明顯下降(P<0.05),GIR明顯升高(P<0.05),與正常對照組水平相噹相(P>0.05);高脂對照組灌胃4週後PPAR-γmRNA錶達水平與正常對照組錶達水平相噹(P>0.05),高脂+紅景天榦預組較正常對照組及高脂對照組比較PPAR-γmRNA錶達水平均有明顯升高(P<0.05)。結論紅景天提取物對大鼠的胰島素牴抗有顯著地改善作用,降低血清TG、TC及FFA明顯降低,調節FPG平衡,降低FINS水平,增加GIR,提高胰島素敏感性,提示其作用機製可能與提高胰島素牴抗大鼠肝髒細胞PPAR-γmRNA錶達有關。
목적:관찰중약홍경천제취물대고지음식유도적이도소저항대서이도소민감성급과양화물매체증식물격활수체-γ( PPAR-γ) mRNA표체적영향。방법장70지Wistar대서수궤분위2조,정상대조조20지,고지모형조50지。정상대조조급여기출사료,고지모형조급여고지사료,균위양4주후2조각수궤선취10지대서행고이도소-정포도당겸협시험판단조모시부성공,학인조모성공후장잉여고지모형조대서수궤분위고지대조조화고지+홍경천간예조,매조20지。고지+홍경천간예조급여홍경천제취물1 g/( kg · d)관위,정상대조조급고지대조조급여등용적0.9%록화납주사액관위,기타사양조건불변,계속위양4주。관찰비교각조대서조모후화관위4주후혈청감유삼지( TG)、혈청총담고순( TC)、유리지방산( FFA)、공복혈당( FPG)급공복혈청이도소( FINS)적함량변화,병기록포도당수주솔( GIR)변화급PPAR-γmRNA적표체정황。결과고지모형조대서조모후여정상대조조비교FPG、FINS명현승고(P<0.05),GIR명현하강(P<0.05);고지대조조관위4주후여정상대조조비교TG、TC급FFA수평균명현승고(P<0.05),고지+홍경천간예조교고지대조조TC、TG급FFA수평균명현하강(P<0.05),여정상대조조수평상당(P>0.05);고지대조조관위4주후여정상대조조비교FPG、FINS명현승고(P<0.05),GIR명현하강(P<0.05),고지+홍경천간예조교고지대조조FPG、FINS수평명현하강(P<0.05),GIR명현승고(P<0.05),여정상대조조수평상당상(P>0.05);고지대조조관위4주후PPAR-γmRNA표체수평여정상대조조표체수평상당(P>0.05),고지+홍경천간예조교정상대조조급고지대조조비교PPAR-γmRNA표체수평균유명현승고(P<0.05)。결론홍경천제취물대대서적이도소저항유현저지개선작용,강저혈청TG、TC급FFA명현강저,조절FPG평형,강저FINS수평,증가GIR,제고이도소민감성,제시기작용궤제가능여제고이도소저항대서간장세포PPAR-γmRNA표체유관。
Objective To observe the effect of Rhodiola rosea extract on insulin sensitivity and the expres-sion of Ppar-γin rats with insulin resistance induced by diets .Methods 70 Wistar rats were randomly divided into two groups, normal control group ( n =20) and hyperlipid model group ( n =50).Normal control group given basic feed, hyperlipid model group was given a high -fat feed, feed four weeks later each line 10 rats were randomly se-lected high insulin -glucose clamps test to determine whether a building is successful , confirmed after the success of the building will be residual fatty model group rats were randomly divided into the hyperlipid control group and hyper -lipid +rhodiola intervention group , each group of 20 rats.Rats in hyperlipid +rhodiola intervention group were giv-en Rhodiola rosea extract (1g/kg· d).Rats in normal control group and hyperlipid control group were given equal a -mount of 0.9%sodium chloride injection by intragastric administration .After four weeks The changes of TG , TC, FFA, FBG and FINS in each group were observed , GIR changes and the expression of Ppar -γmRNA were recorded .Results FBG and FINS of rats in hy-perlipid model group were increased obviously compared with those in normal control group (P <0.05), GIR was deceased (P<0.05).The levels of TG, TC and FFA in hyperlipid model group were increased obviously compared with those in normal control group (P<0.05). The levels of TG , TC and FFA in hyperlipid +rhodiola intervention group were decreased obviously compared with those in hyperlipid model group (P<0.05).The levels of TG, TC and FFA were comparable level with the control group (P>0.05).The level of the expression of Ppar -γmRNA in hyperlipid control group was rather with that in normal control group (P>0.05).The level of the expression of Ppar -γmRNA in hyperlipid +rhodiola intervention group was significantly increased as compared with that in hyperlipid model group and normal control group and con -trol group (P<0.05).Conclusion Rhodiola rosea extract has obvious improvement role on insulin resistance of rats, significantly lower serum TG, TC and FFA, adjust the balance of FBG, lower levels of FINS, increase the GIR, improve insulin sensitivity and its mechanism may be related to increase insulin resistance in rat liver cells ppar -gamma mRNA expression.
