目的:探讨吉西他滨和奥沙利铂对肺癌的耐药性。方法取87例肺癌患者肿瘤组织,并进行体外原代细胞培养,采用Mtt法检测吉西他滨、奥沙利铂对肺癌细胞的抑制率和敏感性。敏感性评价标准为高度敏感(抑制率>70%),中度敏感(抑制率50%~70%),不敏感(抑制率<50%)。结果随着药物浓度的增高,两种药物对肺癌细胞生长抑制率显著增加(P<0.05);随着用药时间的延长,对肺癌细胞抑制率也明显增加(P<0.05)。nsclc对两种药物的敏感性为奥沙利铂>吉西他滨;sclc对两种药物的敏感性为吉西他滨>奥沙利铂。结论随着药物浓度的增高和药物作用时间的延长,吉西他滨和奥沙利铂对肺癌细胞生长抑制率显著增加;奥沙利铂主要对nsclc敏感,而吉西他滨对sclc的敏感性较高;因此对肺癌患者需要针对患者的情况实施个体化的治疗方案,这样有助于提高药物的治疗效果。

목적:탐토길서타빈화오사리박대폐암적내약성。방법취87례폐암환자종류조직,병진행체외원대세포배양,채용Mtt법검측길서타빈、오사리박대폐암세포적억제솔화민감성。민감성평개표준위고도민감(억제솔>70%),중도민감(억제솔50%~70%),불민감(억제솔<50%)。결과수착약물농도적증고,량충약물대폐암세포생장억제솔현저증가(P<0.05);수착용약시간적연장,대폐암세포억제솔야명현증가(P<0.05)。nsclc대량충약물적민감성위오사리박>길서타빈;sclc대량충약물적민감성위길서타빈>오사리박。결론수착약물농도적증고화약물작용시간적연장,길서타빈화오사리박대폐암세포생장억제솔현저증가;오사리박주요대nsclc민감,이길서타빈대sclc적민감성교고;인차대폐암환자수요침대환자적정황실시개체화적치료방안,저양유조우제고약물적치료효과。
Objectiveto explore drug resistance of gemcitabine and oxaliplatin on lung cancer.Methods tumor tissues of 87 cases of patients with lung cancer were prepared for primary cell culture in vitro. Mtt method was used to detect inhibition rate and the sensitivity on lung cancer cells by gemcitabine and oxaliplatin. sensitivity evaluation standard was the highly sensitive (inhibition rate > 70%), moderately sensitive (inhibition rate 50% ~ 50%) and not sensitive (inhibition rate <50%).Results as the drug concentration increased, inhibition rates of lung cancer cell growth by two drugs were increased signiifcantly (P<0.05). With the extension of duration, the inhibition rate of lung cancer cells also increased signiifcantly(P<0.05). the sensitivity of the two drugs on nsclc was oxaliplatin better than gemcitabine. While the sensitivity of the two drugs on sclc was gemcitabine better than oxaliplatin.Conclusion With the increasing of drug concentration and the extension of drug duration, gemcitabine and oxaliplatin both showed increasing inhibition rate on the lung cancer cell growth. nsclc was sensitive to oxaliplatin, and sclcwith higher sensitivity of gemcitabine. therefore, the individualized treatment should be carried out on lung cancer patients aiming at the condition of the patients, thus helps to improve the therapeutic effect of drugs.
