Shi Quan Da Bu Decoction%Primary carcinoma of the liver%Tumor vessel
目的:探讨十全大补汤对小鼠原发性肝癌的生长及血管生成的影响,阐明十全大补汤抑制原发性肝癌生长的机制。方法:构建2种荷瘤小鼠原发性肝肿瘤模型,随机分为生理盐水组、十全大补汤组。分别采用生理盐水及十全大补汤治疗10d,摘除眼球取血,剥离瘤体,并测量其大小和质量。酶联免疫吸附测定法检测治疗后小鼠血清中血管内皮生长因子、血管抑素和内皮抑素水平。链霉菌抗生物素蛋白-过氧化物酶连结免疫组织化学法检测转移瘤微血管密度及细胞增殖。结果:小鼠肝肿瘤模型中,十全大补汤组平均肿瘤体积与生理盐水组比较明显变小(P<0.01),与生理盐水组比较,十全大补汤组肿瘤组织Ki67明显降低(P<0.01),MVD明显降低(P<0.01)。ELISA检测显示,与生理盐水组比较,十全大补汤能够降低血清中VEGF及上调ES的表达,差异有统计学意义(P<0.01)。结论:十全大补汤能明显抑制小鼠原发肝肿瘤生长,明显抑制肿瘤血管生成以及与之相关的血管生成调节因子的作用,并且可抑制肿瘤细胞增殖,促进细胞凋亡。应用十全大补汤对控制肝肿瘤的生长具有重要意义。

목적:탐토십전대보탕대소서원발성간암적생장급혈관생성적영향,천명십전대보탕억제원발성간암생장적궤제。방법:구건2충하류소서원발성간종류모형,수궤분위생리염수조、십전대보탕조。분별채용생리염수급십전대보탕치료10d,적제안구취혈,박리류체,병측량기대소화질량。매련면역흡부측정법검측치료후소서혈청중혈관내피생장인자、혈관억소화내피억소수평。련매균항생물소단백-과양화물매련결면역조직화학법검측전이류미혈관밀도급세포증식。결과:소서간종류모형중,십전대보탕조평균종류체적여생리염수조비교명현변소(P<0.01),여생리염수조비교,십전대보탕조종류조직Ki67명현강저(P<0.01),MVD명현강저(P<0.01)。ELISA검측현시,여생리염수조비교,십전대보탕능구강저혈청중VEGF급상조ES적표체,차이유통계학의의(P<0.01)。결론:십전대보탕능명현억제소서원발간종류생장,명현억제종류혈관생성이급여지상관적혈관생성조절인자적작용,병차가억제종류세포증식,촉진세포조망。응용십전대보탕대공제간종류적생장구유중요의의。
Objective:To explore the influence of Shi Quan Da Bu Decoction on the growth of hepatocellular carcinoma and angiogenesis,and to elucidate the mechanism of Shi Quan Da Bu Decoction inhibiting the growth of hepatocellular carcinoma. Methods:We constructed 2 kinds of tumor bearing mice primary liver tumor model.They were randomLy divided into the saline group,and the Shi Quan Da Bu Decoction group.They were treated respectively with saline and Shi Quan Da Bu Decoction in the treatment of 10 days,and then we removed the eye blood,peeled the tumor,and measured the size and quality.After treatment,we detected the expression of vascular endothelial growth factor in serum of mice,angiostatin and endostatin levels by enzyme-linked immunosorbent assay.We detected metastatic tumor microvessel density and proliferation by streptavidin peroxidase immunohistochemistry and enzyme linked chemical method.Results:In the mouse liver tumor model,in Shi Quan Da Bu Decoction, the average tumor volume was significantly smaller than the normal saline group(P<0.01);compared with the normal saline group, in Shi Quan Da Bu Decoction,the tumor tissue Ki67 was significantly lower(P<0.01);MVD was significantly decreased(P<0.01). ELISA test showed that,compared with the normal saline group,Shi Quan Da Bu Decoction could reduce the expression of VEGF in serum and increase ES expression,and the difference was statistically significant(P<0.01).Conclusion:Shi Quan Da Bu Decoction can obviously inhibit the mouse primary liver tumor growth,inhibit tumor angiogenesis and angiogenesis related regulatory factors,and can inhibit the proliferation of tumor cells,promote cell apoptosis.Application of Shi Quan Da Bu Decoction has an important significance to control liver tumor growth.
