以萱草‘香宝’的花托为外植体,用正交试验法研究了不同激素及其不同浓度对愈伤组织诱导、愈伤组织增殖、不定芽诱导和不定根诱导的影响.结果表明:6-BA是影响‘香宝’愈伤组织诱导的主要因素,各因素影响的主次关系为6-BA>2,4-D>NAA>KT,最适愈伤诱导的培养基为 MS+6-BA 2 mg/L+2,4-D 2 mg/L+NAA 0.3 mg/L,愈伤诱导率为81.28%;6-BA 是影响愈伤增殖的主要因素,各因素影响的主次关系为6-BA>NAA>KT>2,4-D,愈伤组织增殖的最佳配方为 MS+6-BA 1 mg/L +KT 1 mg/L+2,4-D 0.2 mg/L+ NAA 0.05 mg/L,愈伤增值倍数为5.271;GA 是影响不定芽诱导的主要因素,各因素影响的主次关系为 GA>6-BA>NAA,最适不定芽诱导的培养基为 MS+6-BA 1 mg/L+NAA 0.1 mg/L +GA 0.5 mg/L,不定芽诱导率为78.21%;IAA是影响不定根诱导的主要因素,因素影响的主次关系为 IAA>MS>蔗糖>NAA,最适不定根诱导的培养基为1/2 MS+ NAA 0.3 mg/L+蔗糖20 g/L,不定根诱导率为94.4%.

Hemeroallis middendorfii Trautv.et Mey ‘Sweet treasure’was used as explants to study the effect of different hormones and their concentrations on the callus induction,proliferation,adventitious bud and roots induction by orthogonal experiment.The results showed that 6-BA was a key factor on the callus induc-tion,the effect sequence of four hormones was 6-BA>2 ,4-D>NAA>KT,the optimum callus induction medium was MS+6-BA 2 mg/L+2,4-D 2 mg/L+ NAA 0.3 mg/L and the callus induction rate was 81.28%.6-BA was also the main factor influencing callus proliferation,the effect sequence of four hormones was 6-BA>NAA>KT>2,4-D,the optimal callus proliferation medium was MS+6-BA 1 mg/L +KT 1 mg/L+2,4-D 0.2 mg/L+ NAA 0.05 mg/L and the callus proliferation rate was 52.71%.GA was a leading factor on the adventitious bud induc-tion,the effect sequence of four hormones was GA>6-BA>NAA,the optimum medium was MS+6-BA 1 mg/L+NAA 0.1 mg/L+GA 0.5 mg/L,and the adventitious bud induction rate was 78.21%;IAA was a major factor on the adventitious roots induction,the effect sequence of four hormones was IAA>MS>sucrose>NAA ,the opti-mum adventitious roots induction medium was 1/2 MS+ NAA 0.3 mg/L+sucrose 20 g/L,and the adventitious roots induction rate was 7 8 .2 1%.
