目的:观察二脱甲氧基姜黄素对体外培养人宫颈癌Hela细胞Caspase-8的作用。方法:MTT法检测二脱甲氧基姜黄素对体外培养人宫颈癌细胞Hela的抑制作用;分光光度计检测其对Caspase-8酶活性的影响;免疫组化法检测其对人宫颈癌细胞Hela Caspase-8蛋白表达的影响。结果:二脱甲氧基姜黄素对人宫颈癌细胞Hela有明显的抑制作用( P<0.001),且呈剂量关系,与姜黄素有显著差异( P<0.001);二脱甲氧基姜黄素能增强Caspase-8酶活性,增强Caspase-8蛋白的表达。结论:二脱甲氧基姜黄素能激活Caspase-8蛋白活性,这可能也是其体外抑制宫颈癌细胞Hela生长的机理之一。

목적:관찰이탈갑양기강황소대체외배양인궁경암Hela세포Caspase-8적작용。방법:MTT법검측이탈갑양기강황소대체외배양인궁경암세포Hela적억제작용;분광광도계검측기대Caspase-8매활성적영향;면역조화법검측기대인궁경암세포Hela Caspase-8단백표체적영향。결과:이탈갑양기강황소대인궁경암세포Hela유명현적억제작용( P<0.001),차정제량관계,여강황소유현저차이( P<0.001);이탈갑양기강황소능증강Caspase-8매활성,증강Caspase-8단백적표체。결론:이탈갑양기강황소능격활Caspase-8단백활성,저가능야시기체외억제궁경암세포Hela생장적궤리지일。
Objective:To observe the effects of two demethoxylation curcumin on Caspase -8 in Hela cells cultured in vitro .Methods:MTT method was used to determine the inhibition effect of two demethoxylation cur-cumin on human cervical carcinoma cell line Hela cultured in vitro;Caspase -8 activity was detected by spec-trophotometry ;Caspase-8 protein expression of human cervical carcinoma cells Hela was determined by immu-nohistochemistry assay .Results: Two demethoxylation curcumin could significantly inhibit the proliferation of human cervical carcinoma cells Hela (P<0.01), the effect was taken shape of dose -dependentment relation-ship, there was obvious difference to compare with curcumin ( P<0.01);two demethoxylation curcumin could enhance Caspase-8 activity and the expression of Caspase -8 protein in Hela cells.Conclusion:Two deme-thoxylation curcumin could activate Caspase -8 protein, which may be one of the mechanism for inhibiting the proliferation of human cervical carcinoma cells Hela .
