目的:观察绞股蓝总皂苷对高糖培养条件下小鼠永生足细胞nephrin、VEGF( vascular endothelial growth factor ,血管内皮生长因子) mRNA表达的影响。方法:利用体外培养条件性永生小鼠足细胞,高糖干预24小时,模拟糖尿病肾病足细胞损伤,分为正常组、高糖模型组、高渗对照组、绞股蓝治疗组(绞股蓝总皂苷高、中、低剂量组)及缬沙坦对照组,Real Time PCR 法检测足细胞nephrin、VEGF mRNA的表达。结果:绞股蓝总皂苷干预组nephrin mRNA含量显著升高,VEGF mRNA表达有一定程度提高,与模型对照组比较,差异有显著统计学意义( P<0.05或P<0.01)。结论:绞股蓝总皂苷可有效调节肾脏足细胞nephrin、VEGF mRNA表达。

목적:관찰교고람총조감대고당배양조건하소서영생족세포nephrin、VEGF( vascular endothelial growth factor ,혈관내피생장인자) mRNA표체적영향。방법:이용체외배양조건성영생소서족세포,고당간예24소시,모의당뇨병신병족세포손상,분위정상조、고당모형조、고삼대조조、교고람치료조(교고람총조감고、중、저제량조)급힐사탄대조조,Real Time PCR 법검측족세포nephrin、VEGF mRNA적표체。결과:교고람총조감간예조nephrin mRNA함량현저승고,VEGF mRNA표체유일정정도제고,여모형대조조비교,차이유현저통계학의의( P<0.05혹P<0.01)。결론:교고람총조감가유효조절신장족세포nephrin、VEGF mRNA표체。
Objective:To observe the effects of Gypenosides on nephrin , VEGFmRNA expressions of Mice's podocytes in hyperglycemia conditions .Methods: Mice’ s podocytes cultured by high glucose in vitro for 24 hours,to make podocyte injury model of diabetic nephropathy , experiment were divided into normal group , model group, high glucose control group , treatment group ( Gynostemma gypenosides high , middle, low dose group ) and valsartan group , the expressions of nephrin ,VEGF mRNA were determined by Real -Time PCR.Results:The expression of nephrin mRNA significantly increased in Gypenosides intervention group ,VEGFmRNA expres-sion was increased to a certain extent ,to compare with model group ,the difference was statistically significant ( P<0.05 or P<0.01 ).Conclusion: Gypenosides can regulate expressions of nephrin 、VEGF mRNA in renal podocyte .
