Shendi bushen capsule%glomerular sclerosis%BMP-7%CTGF rat
目的:探讨参地补肾胶囊对肾小球硬化大鼠BMP-7、CTGF表达的影响。方法:采用单侧肾脏切除同时尾静脉注射阿霉素的方法建立肾小球硬化大鼠模型,实验分为正常对照组、模型组、西药(洛汀新)对照组、中药(苏黄泄浊丸)组和参地补肾胶囊组,分组给药处理8周。检测各组大鼠24小时尿蛋白定量、血肌酐( SCr)、尿素氮( BUN)、血浆蛋白( ALB);光镜观察大鼠肾脏病理学变化;免疫组化法检测BMP-7、CTGF表达。结果:各治疗组大鼠24小时尿蛋白及血SCr、BUN、ALB较模型组明显改善( P<0.05)。各给药组大鼠肾脏病理变化明显改善。各治疗组均能抑制CTGF的表达( P<0.05),上调BMP-7表达(P<0.05);参地补肾胶囊组作用明显优于两阳性对照组(P<0.05)。结论:参地补肾胶囊对肾小球硬化大鼠肾功能具有保护作用,上调BMP-7表达、抑制CTGF表达可能是其部分作用机制。
目的:探討參地補腎膠囊對腎小毬硬化大鼠BMP-7、CTGF錶達的影響。方法:採用單側腎髒切除同時尾靜脈註射阿黴素的方法建立腎小毬硬化大鼠模型,實驗分為正常對照組、模型組、西藥(洛汀新)對照組、中藥(囌黃洩濁汍)組和參地補腎膠囊組,分組給藥處理8週。檢測各組大鼠24小時尿蛋白定量、血肌酐( SCr)、尿素氮( BUN)、血漿蛋白( ALB);光鏡觀察大鼠腎髒病理學變化;免疫組化法檢測BMP-7、CTGF錶達。結果:各治療組大鼠24小時尿蛋白及血SCr、BUN、ALB較模型組明顯改善( P<0.05)。各給藥組大鼠腎髒病理變化明顯改善。各治療組均能抑製CTGF的錶達( P<0.05),上調BMP-7錶達(P<0.05);參地補腎膠囊組作用明顯優于兩暘性對照組(P<0.05)。結論:參地補腎膠囊對腎小毬硬化大鼠腎功能具有保護作用,上調BMP-7錶達、抑製CTGF錶達可能是其部分作用機製。
목적:탐토삼지보신효낭대신소구경화대서BMP-7、CTGF표체적영향。방법:채용단측신장절제동시미정맥주사아매소적방법건립신소구경화대서모형,실험분위정상대조조、모형조、서약(락정신)대조조、중약(소황설탁환)조화삼지보신효낭조,분조급약처리8주。검측각조대서24소시뇨단백정량、혈기항( SCr)、뇨소담( BUN)、혈장단백( ALB);광경관찰대서신장병이학변화;면역조화법검측BMP-7、CTGF표체。결과:각치료조대서24소시뇨단백급혈SCr、BUN、ALB교모형조명현개선( P<0.05)。각급약조대서신장병리변화명현개선。각치료조균능억제CTGF적표체( P<0.05),상조BMP-7표체(P<0.05);삼지보신효낭조작용명현우우량양성대조조(P<0.05)。결론:삼지보신효낭대신소구경화대서신공능구유보호작용,상조BMP-7표체、억제CTGF표체가능시기부분작용궤제。
Objective:To explore the effects of Shendi bushen capsule on BMP-7、CTGF expressions in rats with glomerular sclerosis.Methods:Unilateral nephrectomy and injection of adriamycin by tail vein to establish the model of glomerular sclerosis.SD rats were randomly divided into normal control group , model group, western medi-cine (lotensin) group,Chinese medicine (Suhuang xiezhuo pill) group and Shendi bushen capsule group,each group was given the corresponding medicine for 8 weeks.rats’24hours urinary protein,serum creatinine(SCr)、urea nitrogen (BUN)、plasma albumin(ALB)were determined;the pathological changes of rats’kidney were observed by light micro-scope;the expressions of BMP-7、CTGF in renal tissue was determined by immunohistochemical method.Results:To compare with model group,24 hours urinary protein and blood SCr、BUN and ALB were improved significantly in each treatment group(P<0.05).the pathologic structure changes of renal glomerular, tubules, interstitial and vascular were improved obviously in each treatment group.The expression of CTGF was inhibit and expression of BMP-7 was up-regulated in each treatment group, the effects of Shendi bushen capsule group were the most obvious(P<0.05).Conclusion:Shendi bushen capsule can protect rats’ renal function ,its mechanism may be related to increasing the expression of BMP-7 and inhibiting CTGF expression.
