目的:观察舒心稳斑颗粒对实验兔AS斑块稳定性及MMP-9、I型胶原蛋白表达的影响。方法:运用高脂高胆固醇饲料喂养12周制备日本大耳白兔动脉粥样硬化模型,用舒心稳斑颗粒灌胃给药治疗4周,采用双抗体夹心酶联免疫吸附法检测血清中I型胶原蛋白的含量;采用免疫组化法分析I型胶原蛋白、MMP-9在主动脉粥样斑块中的表达;同时观察主动脉粥样硬化斑块的病理学改变。结果:与模型组比较,舒心稳斑颗粒能升高血清中Ⅰ型胶原蛋白的含量,差异具有显著统计学意义( P<0.01);同时能降低斑块中MMP-9的表达水平,升高Ⅰ型胶原蛋白在斑块中的表达水平。病理学检查结果显示,采用舒心稳斑颗粒干预后,主动脉粥样硬化斑块的病理学形态、结构均发生改变,斑块结构更加趋于稳定。结论:舒心稳斑颗粒可能通过抑制斑块内炎症反应,调节动脉粥样硬化斑块纤维帽中的胶原纤维降解与合成失衡状态,进而对AS斑块起稳定作用。

목적:관찰서심은반과립대실험토AS반괴은정성급MMP-9、I형효원단백표체적영향。방법:운용고지고담고순사료위양12주제비일본대이백토동맥죽양경화모형,용서심은반과립관위급약치료4주,채용쌍항체협심매련면역흡부법검측혈청중I형효원단백적함량;채용면역조화법분석I형효원단백、MMP-9재주동맥죽양반괴중적표체;동시관찰주동맥죽양경화반괴적병이학개변。결과:여모형조비교,서심은반과립능승고혈청중Ⅰ형효원단백적함량,차이구유현저통계학의의( P<0.01);동시능강저반괴중MMP-9적표체수평,승고Ⅰ형효원단백재반괴중적표체수평。병이학검사결과현시,채용서심은반과립간예후,주동맥죽양경화반괴적병이학형태、결구균발생개변,반괴결구경가추우은정。결론:서심은반과립가능통과억제반괴내염증반응,조절동맥죽양경화반괴섬유모중적효원섬유강해여합성실형상태,진이대AS반괴기은정작용。
Objective:To observe the effects of Shuxin wenban granule on the stability of AS plaque and the expressions of MMP-9 and COL-l in rabbits.Methods: Using the high cholesterol diet to induce AS rabbit models for 12 weeks, then, giving Shuxin wenban granule for 4 weeks, finally,detecting the serum levels of COL-I in the rabbits by double antibody sandwich enzyme -linked immunosorbent method and the surface density of MMP-9 and COL-I in atherosclerotic plaques by immunohistochemical staining .At the same time , observing the pathological changes of aorta in atherosclerotic rabbits .Results:To compared with model group , Shuxin wen-ban granule could increase the serum levels of COL -I,the difference was statistically significant (P<0.01);similarly, Shuxin wenban granule could significantly decrease the surface density of MMP -9 and increase the surface density of COL-I in atherosclerotic plaques,the difference was statistically significant (P<0.01).Pa-thology showed that Shuxin wenban granule changed the pathological morphology , structure of atherosclerotic plaque , caused the plaque to stabler .Conclusion: Shuxin wenban granules could stabilize atherosclerotic plaques by inhibiting the inflammatory response of atherosclerotic plaque and affecting the imbalance of collagen fibers’ degradation and synthesis in atherosclerotic plaque fibrous cap .
