Tetramethylpyrazine%sperm motility%invitro
目的:研究不同浓度的川芎嗪注射液(TMP)对体外人精子运动参数的影响,并初步探讨其可能的作用机制。方法20例健康生育男性手淫供精,将上游优化处理的精子,与不同浓度TMP注射液共同孵育10、30、60min,采用计算机辅助精子分析系统观察不同浓度TMP(0.1、1、10mmol/L)体外对精子各运动参数的作用。结果与对照组相比,1mmol/L浓度组,除60min时精子活率[(a+b+c)级精子%]未下降外,其余时间精子活率、前向运动精子百分率[(a+b)级精子%]、平均直线速度(VSL)和平均路径速度(VAP)等观察指标均明显下降,差异均具统计学意义(P<0.05,P<0.01)。10mmol/L浓度组,精子活率、前向运动精子百分率、VSL和VAP等观察指标也均明显下降,差异均有统计学意义(P<0.01)。与对照组相比,侧摆幅度(ALH)除0.1mmol/L浓度组和1mmol/L浓度组30min时未下降外,余均有下降,但差异无统计学意义(P>0.05)。结论川芎嗪体外能显著降低精子运动能力,为男性节育提供了一种新选择。

Objective To investigate the effects of the Tetramethylpyrazine (TMP) on human sperm motility in vitro and further explore its possible mechanisms. Methods Sperm samples from 20 healthy fertile men were collected by masturbation, and then they were treated by swim-up technique to produce a spermatozoa solution of high motility. Subsequently they were incubated with different concentration of tetramethylpyrazine solution( 0.1, 1, 10mmol/L) invitro. Sperm motility parameters at incubation time of 10, 30 and 60min were measured by computer-aided sperm analysis (CASA). Results Contrast to the control group, In group 1mmol/L and 10 mmol/L,TMP markedly decreased the sperm live rate, progressive motility, VSL, VAP, there were significant difference statistically contrast to the control group. But ALH had no significant difference statistically contrast to the control group. Conclusion The data suggest that tetramethylpyrazine can inhibit sperm motility in vitro, which may be a new approach for male contraception.
