目的:观察溶血磷脂酰胆碱(Lysophosphatidyl choline,LPC)对人脐静脉内皮细胞(Human umbilical vein endothelial cells,HUVECs)血管紧张素系统的影响。方法体外培养HUVECs,分为正常对照组、低浓度(10μg/L)LPC组、中浓度(20μg/L)LPC组和高浓度(40μg/L)LPC组。采用放射免疫法和RT-PCR法检测LPC对HUVECs血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)蛋白及AngⅡ1型受体(An-giotensinⅡtype1 receptor,AT1R)、AngⅡ2型受体(AngⅡtype2 receptor,AT2R)mRNA表达的影响。结果与正常对照组相比,LPC可显著上调HUVECs AngⅡ蛋白及AT1R mRNA的表达。结论 LPC能够上调人脐静脉内皮细胞血管紧张素系统部分组分的表达。

목적:관찰용혈린지선담감(Lysophosphatidyl choline,LPC)대인제정맥내피세포(Human umbilical vein endothelial cells,HUVECs)혈관긴장소계통적영향。방법체외배양HUVECs,분위정상대조조、저농도(10μg/L)LPC조、중농도(20μg/L)LPC조화고농도(40μg/L)LPC조。채용방사면역법화RT-PCR법검측LPC대HUVECs혈관긴장소Ⅱ(AngiotensinⅡ,AngⅡ)단백급AngⅡ1형수체(An-giotensinⅡtype1 receptor,AT1R)、AngⅡ2형수체(AngⅡtype2 receptor,AT2R)mRNA표체적영향。결과여정상대조조상비,LPC가현저상조HUVECs AngⅡ단백급AT1R mRNA적표체。결론 LPC능구상조인제정맥내피세포혈관긴장소계통부분조분적표체。
Objective To observe the effect of Lysophosphatidyl choline (LPC) on Renin-angiotensin system (RAS) of human umbilical vein endothelial cells (HUVECs). Methods HUVECs were cultured in vi-tro. The study was designated to 4 groups: (1) normal control, (2) low-concentration LPC group (10μg/L), (3) middle-concentration LPC group (20μg/L), (4) high-concentration LPC group(40μg/L). Expression of angiotensinⅡ (AngⅡ ) and AngⅡ type1 receptor(AT1R), AngⅡ type2 receptor(AT2R) mRNA were ob-served by radio-immunity and RT-PCR, respectively. Results Compared with normal control group, LPC could significantly up-regulate the expression of AngⅡ and AT1R mRNA. Conclusion LPC could partly up-regulate the expression of RSA of human umbilical vein endothelial cells.
