目的:通过研究RhoA、RhoB和RhoC在肺癌组织及细胞系中表达水平的变化,探讨Rho亚家族与肺癌发生、发展及转移的关系。方法应用荧光定量RT-PCR法、Western blotting法研究四种肺癌细胞系RhoA、RhoB和RhoC mRNA和蛋白表达水平变化,免疫组织化学法检测45例人肺癌组织RhoA、RhoB和RhoC蛋白表达。结果四种肺癌细胞系RhoA和RhoB mRNA和蛋白表达水平显著高于对照组(P均<0.05),肺癌细胞系组间比较,表达水平差异无统计学意义(P均>0.05),各组间RhoC mRNA和蛋白表达水平差异无统计学意义(P均>0.05)。RhoA、RhoB蛋白在人肺癌组织呈阳性表达,中分化鳞癌组阳性率与非中分化鳞癌组阳性率差异无统计学意义(P均>0.05),而RhoC全部呈阴性表达。结论 RhoA、RhoB在肺癌组织中异常高表达,提示Rho亚家族参与了肺癌的发生、分化和转移过程。

목적:통과연구RhoA、RhoB화RhoC재폐암조직급세포계중표체수평적변화,탐토Rho아가족여폐암발생、발전급전이적관계。방법응용형광정량RT-PCR법、Western blotting법연구사충폐암세포계RhoA、RhoB화RhoC mRNA화단백표체수평변화,면역조직화학법검측45례인폐암조직RhoA、RhoB화RhoC단백표체。결과사충폐암세포계RhoA화RhoB mRNA화단백표체수평현저고우대조조(P균<0.05),폐암세포계조간비교,표체수평차이무통계학의의(P균>0.05),각조간RhoC mRNA화단백표체수평차이무통계학의의(P균>0.05)。RhoA、RhoB단백재인폐암조직정양성표체,중분화린암조양성솔여비중분화린암조양성솔차이무통계학의의(P균>0.05),이RhoC전부정음성표체。결론 RhoA、RhoB재폐암조직중이상고표체,제시Rho아가족삼여료폐암적발생、분화화전이과정。
To investigate changes of expressions of RhoA and RhoB and RhoC in lung cancer tissues to explore the relationship of Rho subfamily with development,migration,and metastasis of lung carcinoma. Methods The expressions of RhoA,RhoB and RhoC mRNA and protein in four different lung cancer cell lines (LTEP-a-2,SK-MES-1,NCI-H446,and NCI-H292) were detected by real-time quantitative reverse transcription polymerase chain reaction(QRT-PCR) and Western blotting, and were compared with MRC-5(control group). The expressions of RhoA,RhoB and RhoC protein in 45 cases of lung cancer tissue by immunocytochemistry. Results The expressions of RhoA and RhoB mRNA and protein in lung cancer cell line LTEP-a-2,SK-MES-1,NCI-H446,and NCI-H292 were significantly higher than those in control group (all P<0.05),while no statistical difference was found among four lung cancer cell lines(all P>0.05). The expressions of RhoA and RhoB protein in human lung cancer tissue were positive,but the levels of them were not statistically different between moderately differentiated squamous carcinoma group and non-moderately differentiated squamous carcinoma group (all P>0.05). The expression of RhoC protein was negative. Conclusion The expressions of RhoA and RhoB were elevated in lung cancer,which indicates that Rho subfamily involve in the development,migration and metastasis of lung cancer.
