目的探讨促睡眠调节中枢下丘脑腹外侧视前区(VLPO)与促觉醒调节中枢结节乳头体核(TMN)之间是否具有双向调节的直接通路。方法将SD大鼠随机分为对照组( ACSF 组)与实验组( TMN + ACSF&VLPO + DiO 组, TMN+DiO&VLPO+ACSF组),采用脑立体定位技术,核团内微量注射、冰冻切片等方法观察和记录大鼠VLPO和TMN分别注射细胞膜荧光探针Fast DiO后的TMN和VLPO的荧光信号。结果 TMN+ACSF&VLPO+DiO组大鼠注射Fast DiO术后72 h, TMN 脑区神经细胞可见明显的绿色荧光;TMN+DiO&VLPO+ACSF组大鼠注射Fast DiO术后72 h, VLPO脑区神经细胞也可见明显的绿色荧光。结论 VLPO与TMN间有双向直接神经纤维投射。
目的探討促睡眠調節中樞下丘腦腹外側視前區(VLPO)與促覺醒調節中樞結節乳頭體覈(TMN)之間是否具有雙嚮調節的直接通路。方法將SD大鼠隨機分為對照組( ACSF 組)與實驗組( TMN + ACSF&VLPO + DiO 組, TMN+DiO&VLPO+ACSF組),採用腦立體定位技術,覈糰內微量註射、冰凍切片等方法觀察和記錄大鼠VLPO和TMN分彆註射細胞膜熒光探針Fast DiO後的TMN和VLPO的熒光信號。結果 TMN+ACSF&VLPO+DiO組大鼠註射Fast DiO術後72 h, TMN 腦區神經細胞可見明顯的綠色熒光;TMN+DiO&VLPO+ACSF組大鼠註射Fast DiO術後72 h, VLPO腦區神經細胞也可見明顯的綠色熒光。結論 VLPO與TMN間有雙嚮直接神經纖維投射。
목적탐토촉수면조절중추하구뇌복외측시전구(VLPO)여촉각성조절중추결절유두체핵(TMN)지간시부구유쌍향조절적직접통로。방법장SD대서수궤분위대조조( ACSF 조)여실험조( TMN + ACSF&VLPO + DiO 조, TMN+DiO&VLPO+ACSF조),채용뇌입체정위기술,핵단내미량주사、빙동절편등방법관찰화기록대서VLPO화TMN분별주사세포막형광탐침Fast DiO후적TMN화VLPO적형광신호。결과 TMN+ACSF&VLPO+DiO조대서주사Fast DiO술후72 h, TMN 뇌구신경세포가견명현적록색형광;TMN+DiO&VLPO+ACSF조대서주사Fast DiO술후72 h, VLPO뇌구신경세포야가견명현적록색형광。결론 VLPO여TMN간유쌍향직접신경섬유투사。
Objective To investigate the research of the direct nerve fiber projections between ventrolateral preop-tic nucleus ( VLPO) and tuberomammillary nucleus ( TMN) in rats. Methods SD rats were randomly divided into control group ( ACSF group) and experimental groups ( TMN+ACSF&VLPO+DiO group and TMN+DiO&VLPO+ACSF group) . Brain stereotaxic technique, microinjection and frozen section technique were used to detect and record the fluorescence signal in TMN and VLPO after injecting fluorescence dye, Fast DiO, into VLPO and TMN, respectively. Results In TMN+ACSF&VLPO+DiO group, the green fluorescence signals in TMN could be de-tected after injected Fast DiO dye 72 h. Meanwhile, in TMN+DiO&VLPO+ACS group, the green fluorescence signals could be detected in VLPO after injected Fast DiO dye 72 h. Conclusion There have bi-directional nerve fiber projection between VLPO and TMN.
