硒与硫化氢生成酶均在动物生理活动中起重要作用。为探讨硒缺乏对鸡内源性硫化氢生成酶的影响,在建立鸡低硒模型后,分别于15、25、35、45、55、65 d采集肝组织,应用实时荧光定量PCR法检测H2S生成酶(CSE、CBS、3-MST)mRNA表达量。结果表明,低硒可诱导CSE、CBS和3-MST mRNA表达量升高,但无时间效应,揭示H2S生成酶在硒缺乏导致的损伤中有重要作用。制备CSE多克隆抗体,为今后CSE蛋白水平研究奠定基础。

서여류화경생성매균재동물생리활동중기중요작용。위탐토서결핍대계내원성류화경생성매적영향,재건립계저서모형후,분별우15、25、35、45、55、65 d채집간조직,응용실시형광정량PCR법검측H2S생성매(CSE、CBS、3-MST)mRNA표체량。결과표명,저서가유도CSE、CBS화3-MST mRNA표체량승고,단무시간효응,게시H2S생성매재서결핍도치적손상중유중요작용。제비CSE다극륭항체,위금후CSE단백수평연구전정기출。
Both selenium and sulfide-producing enzyme play important roles in the physiological activity of the animals. The aim of this investigate revealed the effect of generate hydrogen sulfide enzyme induced by the chicken model of selenium deficiency. Liver tissue were col ected on 15, 25, 45, 55, 65 d. Real-time PCR was used to detect the expression of mRNA H2S producing enzyme (CSE, CBS, 3-MST). The results showed that the expression level of selenium could induce the mRNA in CSE, CBS and 3-MST, but there was no effect of time, suggesting that H2S generation enzymes played an important role in the injury in selenium deficiency. At the same time, the preparation of CSE polyclonal antibody was prepared, providing a reference research on the level of CSE protein in the future.
