目的:探讨异基因骨髓间充质干细胞(BMSC)与外周血单个核细胞(PBMC)在体外共培养的相互影响。方法将异基因成人来源的BMSC与PBMC在体外分别按1:10混合共培养,采用氚胸腺嘧啶核甙掺入方法检测PHA刺激PBMC增殖的活性,采用ELISA方法检测培养液中分泌的IL-6、IL-8和PGE2水平。结果 BMSC与PBMC共培养组的PBMC增殖率明显减少(P<0.01),抑制率均超过50%。加入PBMC共培养后,BMSC分泌IL-6、IL-8和PGE2显著增加(P<0.01)。结论 BMSC可以抑制PHA刺激引起的PBMC增殖,PBMC可以促进BMSC分泌免疫活性因子,BMSC可能具有双向的免疫调控作用。

목적:탐토이기인골수간충질간세포(BMSC)여외주혈단개핵세포(PBMC)재체외공배양적상호영향。방법장이기인성인래원적BMSC여PBMC재체외분별안1:10혼합공배양,채용천흉선밀정핵대참입방법검측PHA자격PBMC증식적활성,채용ELISA방법검측배양액중분비적IL-6、IL-8화PGE2수평。결과 BMSC여PBMC공배양조적PBMC증식솔명현감소(P<0.01),억제솔균초과50%。가입PBMC공배양후,BMSC분비IL-6、IL-8화PGE2현저증가(P<0.01)。결론 BMSC가이억제PHA자격인기적PBMC증식,PBMC가이촉진BMSC분비면역활성인자,BMSC가능구유쌍향적면역조공작용。
Objective To explore the interaction of allogeneic human bone marrow mesenchymal stem cell (BMSC) and peripheral blood mononuclear cell (PBMC) cocultured in vitro. Methods Human BMSC and PBMC isolated from unrelated donors were cocultured (BMSC to PBMC ratio, 1:10). 3H-TdR incorporation test were used to evaluate PHA-induced proliferation. ELISA test was used to determine the levels of IL-6, IL-8 and PGE2 in cell cul-ture supernatant. Results The presence of BMSC resulted in a statistically significant (P<0.01) decrease in PHA-in-duced proliferation in PBMC and the inhibition rate was more than 50%. The levels of IL-6, IL-8 and PGE2 signifi-cantly (P<0.01) increased when PBMC were added to BMSC. Conclusion BMSC can mediate suppression of PHA-induced PBMC proliferation. PBMC can promote BMSC to secrete positive and negative immune factors. BM-SC may have dual effects on immune reactions.
