目的:观察知母总皂苷对Aβ25~35诱导PC12细胞凋亡及凋亡相关基因Bax表达的影响。方法培养PC12细胞,实验分为6组:正常组、模型组、知母低剂量组、知母中剂量组、知母高剂量组、西药组。20μmol/L Aβ25~35作用PC12细胞48 h,诱导其凋亡。采用流式细胞仪检测各组细胞凋亡率的变化;采用RT-PCR检测各组细胞中Bax mRNA表达水平的变化;采用 Western印迹技术检测各组细胞中 Bax蛋白的表达水平的变化。结果和模型组比较,知母总皂苷能显著降低Aβ25~35诱导的凋亡(P<0.05),能显著性降低凋亡基因Bax mRNA 和蛋白的表达水平(P<0.05)。结论知母总皂苷能降低Aβ25~35诱导的PC12细胞凋亡率,其机制可能是降Bax mRNA及蛋白表达水平。

목적:관찰지모총조감대Aβ25~35유도PC12세포조망급조망상관기인Bax표체적영향。방법배양PC12세포,실험분위6조:정상조、모형조、지모저제량조、지모중제량조、지모고제량조、서약조。20μmol/L Aβ25~35작용PC12세포48 h,유도기조망。채용류식세포의검측각조세포조망솔적변화;채용RT-PCR검측각조세포중Bax mRNA표체수평적변화;채용 Western인적기술검측각조세포중 Bax단백적표체수평적변화。결과화모형조비교,지모총조감능현저강저Aβ25~35유도적조망(P<0.05),능현저성강저조망기인Bax mRNA 화단백적표체수평(P<0.05)。결론지모총조감능강저Aβ25~35유도적PC12세포조망솔,기궤제가능시강Bax mRNA급단백표체수평。
Objective To observe the effects of Anemarrhena total saponins on beta-amyloid ( Aβ)-induced PC12 cell apoptosis and the mRNA and protein expression of Bax.Methods PC12 cells was stimulated by Aβ25~35( 20μmol/L)for 48 h.Experimental groups were normal, model, Anemarrhena total saponins (5 mg/L, 10 mg/L and 20 mg/L) and the donepezil HCl groups.The apoptosis rate was ana-lyzed by fluorescence microscopy.The expression level of the Bax gene was detected by using semi-quantitative reverse transcriptase PCR ( RT-PCR).The expression levels of Bax protein was assayed by western blot.Results Anemarrhena total saponins markedly attenuated Aβ-inducing injury in PC12 cells, compared with model group (P<0.05),and it dramatically reduced the expression levels of pro-apoptotic gene Bax mRNA and protein , compared with those of model group ( P<0.05 ).Conclusions Anemarrhena total saponins treatment decrease Aβ25~35-induced the apoptosis rate of PC 12 cells, and it decreases the expression levels of Bax mRNA and protein.
