目的:观察不同针刺刺激量对寒凝证类痛经大鼠扭体反应和子宫前列腺素E2(PGE2)、前列腺素F2α(PGF2α)含量的影响,初步探讨针刺的量效关系。方法:将雌性大鼠40只,随机分为正常组、模型组、刺激量A组(粗针进针深,针刺时间长)、刺激量B组(细针进针浅,针刺时间短)。除正常组外,模型组、刺激量A组和刺激量B组采用全身冷冻法结合苯甲酸雌二醇注射法造模。模型制备后,针刺治疗20 min,观察大鼠的扭体反应,并测定大鼠子宫组织内前列腺素E2(PGE2)、前列腺素F2α(PGF2α)含量。结果:①与正常组比较,模型组扭体潜伏期明显缩短(P<0.01),扭体次数及扭体评分明显增加(P<0.01)。与模型组比较,刺激量A组扭体潜伏期明显延长(P<0.05),扭体次数及扭体评分明显减少(P<0.01);刺激量B组扭体次数明显减少(P<0.01)、扭体评分明显减少(P<0.05)。②与正常组比较,模型组PGF2α含量、PGF2α/PGE2比值明显升高(P<0.01)。与模型组比较,刺激量A组PGF2α含量、PGF2α/PGE2比值明显降低(P<0.01);刺激量 B 组 PGF2α含量明显降低(P<0.05),PGE2含量明显升高(P<0.01), PGF2α/PGE2比值明显降低(P<0.01)。结论:针刺刺激量与镇痛效应具有相关性。针刺可明显减轻大鼠的类痛经反应,可能的机制是通过调节子宫前列腺素E2(PGE2)、前列腺素F2α(PGF2α)的紊乱水平而缓解子宫痉挛性收缩。

목적:관찰불동침자자격량대한응증류통경대서뉴체반응화자궁전렬선소E2(PGE2)、전렬선소F2α(PGF2α)함량적영향,초보탐토침자적량효관계。방법:장자성대서40지,수궤분위정상조、모형조、자격량A조(조침진침심,침자시간장)、자격량B조(세침진침천,침자시간단)。제정상조외,모형조、자격량A조화자격량B조채용전신냉동법결합분갑산자이순주사법조모。모형제비후,침자치료20 min,관찰대서적뉴체반응,병측정대서자궁조직내전렬선소E2(PGE2)、전렬선소F2α(PGF2α)함량。결과:①여정상조비교,모형조뉴체잠복기명현축단(P<0.01),뉴체차수급뉴체평분명현증가(P<0.01)。여모형조비교,자격량A조뉴체잠복기명현연장(P<0.05),뉴체차수급뉴체평분명현감소(P<0.01);자격량B조뉴체차수명현감소(P<0.01)、뉴체평분명현감소(P<0.05)。②여정상조비교,모형조PGF2α함량、PGF2α/PGE2비치명현승고(P<0.01)。여모형조비교,자격량A조PGF2α함량、PGF2α/PGE2비치명현강저(P<0.01);자격량 B 조 PGF2α함량명현강저(P<0.05),PGE2함량명현승고(P<0.01), PGF2α/PGE2비치명현강저(P<0.01)。결론:침자자격량여진통효응구유상관성。침자가명현감경대서적류통경반응,가능적궤제시통과조절자궁전렬선소E2(PGE2)、전렬선소F2α(PGF2α)적문란수평이완해자궁경련성수축。
Objective:To observe the effects of different quantity of acupuncture stimulation on writhing re-sponse and uterine prostaglandin E2(PGE2),prostaglandin F2α(PGF2α) contents in dysmenorrheal rats with cold stagnation syndrome,to study relationship between quantity and effect of acupuncture stimulating. Methods:Total 40 female rats were divided into 4 groups randamly,including:normal group,model group,A stimuli group,B stimuli group. Except for normal group,the models were established by general freezing method combined with estradiol benzoate injection molding. After model preparation ,acupuncture treatment were giv-en for 20 min,to observe the writhing response in rats,and to detect PGE2,PGF2α content in rat uterus tis sue. Results:①Compared with the normal group,the writhing latency of the model group was significantly shortened(P<0.01),the writhing times and writhing score increased significantly(P<0.01). Compared with the model group,writhing latency of A stimuli group significantly longthened(P<0.05),the writhing times and writhing score decreased significantly(P<0.01). B stimuli group,the writhing times decreased signif-icantly(P<0.05),the writhing score decreased significantly(P<0.01). ②Compared with the normal group,PGF2αcontent,PGF2α/PGE2 of the model group increased significantly(P<0.01). Compared with the model group, PGF2α content,PGF2α/PGE2 of A stimuli group decreased significantly(P<0.01). In B stimuli group,PGF2αcontent decreased significantly(P<0.05),PGE2 content increased significantly(P<0.01),PGF2α/PGE2 decreased significantly(P<0.01). Conclusion:Acupuncture can obviously reduce pain reactions in dysmenorrheal rats, relieve spastic contraction of the uterus by regulating disorder level of uterine PGE2,PGF2α,which confirmed the correlation between acupuncture stimulating quantity and analgesic effect.
