目的:探讨当归补血汤含药血清对心衰后心肌能量代谢的影响。方法:体外培养心肌细胞,实验分为空白对照组、血管紧张素Ⅱ(AngⅡ)组、AngⅡ+缬沙坦组、AngⅡ+含药血清组;分别用塞唑蓝(MTT)法检测各组心肌细胞总蛋白含量和细胞活力;酶联免疫法检测乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)活性;激光共聚焦显微镜观察线粒体膜电位(MMP)变化。结果:与空白对照组比较,AngⅡ组心肌细胞总蛋白含量、细胞活力、LDH、SDH活性及MMP显著升高(P<0.05);与AngⅡ组比较,AngⅡ+含药血清组心肌细胞总蛋白含量、细胞活力、LDH、SDH活性及MMP显著降低(P<0.05)。结论:当归补血汤含药血清能通过抑制肥大心肌细胞LDH、SDH活性及MMP的病理性升高起到改善心肌能量代谢障碍,保护心肌细胞的作用。

목적:탐토당귀보혈탕함약혈청대심쇠후심기능량대사적영향。방법:체외배양심기세포,실험분위공백대조조、혈관긴장소Ⅱ(AngⅡ)조、AngⅡ+힐사탄조、AngⅡ+함약혈청조;분별용새서람(MTT)법검측각조심기세포총단백함량화세포활력;매련면역법검측유산탈경매(LDH)、호박산탈경매(SDH)활성;격광공취초현미경관찰선립체막전위(MMP)변화。결과:여공백대조조비교,AngⅡ조심기세포총단백함량、세포활력、LDH、SDH활성급MMP현저승고(P<0.05);여AngⅡ조비교,AngⅡ+함약혈청조심기세포총단백함량、세포활력、LDH、SDH활성급MMP현저강저(P<0.05)。결론:당귀보혈탕함약혈청능통과억제비대심기세포LDH、SDH활성급MMP적병이성승고기도개선심기능량대사장애,보호심기세포적작용。
Objective:To explore the effect of serum containing Danggui Buxue decoction on the myocardial energy metabolism after heart failure. Methods:Myocardial cells were cultured in vitro,the cardiomyocytes were divided into 4 groups:control group,AngⅡ group,AngⅡ+ valsartan group and Ang Ⅱ+ medicated serum group. The cardiomyocytes activity and total protein content were measured by MTT method. The ac-tivities of lactate dehydrogenase(LDH),succinate dehydrogenase(SDH) were detected by ELISA. Laser con-focal scanning microscopy was used to detect the change of MMP. Results:In AngⅡgroup,the cardiomy-ocytes activity,total protein content,activities of SDH and LDH and the MMP change were significantly higher than those of the control group(P<0.05). In AngⅡ+ medicated serum group,the cardiomyocytes ac-tivity,total protein content,activities of SDH and LDH and the MMP change were significantly lower than those of the AngⅡ group(P<0.05). Conclusion:Serum containing Danggui Buxue decoction could ameliorate myocardial energy metabolism and prevent hypertrophic cardiomyocytes from injuring ,the mechanism may be related to restrain the activity of LDH,SDH and MMP of myocardial hypertrophy.
