目的:探讨乳腺癌耐药细胞株中P-gp、CD147和E-cadherin表达的意义。方法采用Western blot 方法,比较不耐药细胞、药物诱导的耐药细胞和mdr1基因导入的耐药细胞中P-gp、CD147和E-cadherin(E-cd)表达的差异性。结果 P-gp在不耐药细胞中表达很弱,而在两株耐药细胞中均呈高表达。 CD147在药物诱导的耐药细胞中表达增强,分子量较小。而E-cd在药物诱导的耐药细胞中表达缺失。结论药物诱导和基因转入均能造成乳腺癌细胞产生多药耐药,而CD147和E-cd在三株乳腺癌细胞中的表达差异与mdr1基因或P-gp蛋白表达并无直接关系,其内在的关系及临床意义值得深入探讨。

목적:탐토유선암내약세포주중P-gp、CD147화E-cadherin표체적의의。방법채용Western blot 방법,비교불내약세포、약물유도적내약세포화mdr1기인도입적내약세포중P-gp、CD147화E-cadherin(E-cd)표체적차이성。결과 P-gp재불내약세포중표체흔약,이재량주내약세포중균정고표체。 CD147재약물유도적내약세포중표체증강,분자량교소。이E-cd재약물유도적내약세포중표체결실。결론약물유도화기인전입균능조성유선암세포산생다약내약,이CD147화E-cd재삼주유선암세포중적표체차이여mdr1기인혹P-gp단백표체병무직접관계,기내재적관계급림상의의치득심입탐토。
Objective To explore the expression of P-gp、CD147 and E-cadherin in drug-resistant breast cancer cells . Methods The expression of P-gp,CD147 and E-cd in multi-drug resistance ( MDR) human breast cancer cell line MCF 7/Adr ( induced by adriamycin ) , MCF7/MDR ( mdr1 gene transmited ) and their sensitive parental line ( MCF7 ) were compared by Western blot.Results The expression of P-gp was much higher in MCF7/Adr and MCF7/MDR than that of MCF7.Increased production of CD147,with ligher molecular weight ,was observed in the MCF7/Adr.The expression of E-cd could be find in MCF7/MDR and MCF7,while lost in MCF7/Adr.Conclusion Both adriamycin and gene transmition can induce breast cancer cell to produce MDR.The expression of CD147 and E-cd in the 3 cancer cell lines has no relation with mdr 1 gene or expression of P-gp,their relationship is worthy of further investigation .
