目的:检测人大肠癌组织中Lumican基因及蛋白的表达,探讨其与大肠癌患者临床病理指标及预后之间的关系。方法:收集外科手术切除并经病理证实的27例新鲜大肠癌组织标本,以同一患者远离肿瘤的正常大肠组织为对照,采用实时荧光定量PCR法检测Lumican基因表达,剩余组织采用石蜡包埋用于免疫组化检测Lumican蛋白。另从我院病理科收集同期37例大肠癌组织蜡块,采用免疫组化SP法共检测64例大肠癌组织、27例正常大肠黏膜组织中Lumican蛋白表达。结果:27例新鲜大肠癌组织及对应正常大肠组织中Lumican的表达有显著差异性( P<0.001),Lumican mRNA在大肠癌组织表达明显增高[以正常组织为1,癌组织Lumican基因表达上调(5.35±0.66)倍]。64例大肠癌组织Lumican蛋白阳性表达率为73.44%(47/64),显著高于远离肿瘤的正常组织表达率0%(0/27)(P<0.05)。Lumican表达与肿瘤的病理分级、Dukes分期、淋巴结是否转移有关(P<0.001),而与患者性别、年龄、病变部位无关(P>0.05)。27例大肠癌标本Lu-mican蛋白量与mRNA相对表达量呈正相关(r=0.123),但无统计学意义(P>0.05)。结论:人大肠癌组织Lumican mRNA及蛋白均表达升高,Lumican高表达与大肠癌的临床病理特征及侵袭、转移特性密切相关,在大肠癌发生、发展过程可能起着重要作用。Lumican可作为判断大肠癌生物学行为的重要指标之一。

목적:검측인대장암조직중Lumican기인급단백적표체,탐토기여대장암환자림상병리지표급예후지간적관계。방법:수집외과수술절제병경병리증실적27례신선대장암조직표본,이동일환자원리종류적정상대장조직위대조,채용실시형광정량PCR법검측Lumican기인표체,잉여조직채용석사포매용우면역조화검측Lumican단백。령종아원병이과수집동기37례대장암조직사괴,채용면역조화SP법공검측64례대장암조직、27례정상대장점막조직중Lumican단백표체。결과:27례신선대장암조직급대응정상대장조직중Lumican적표체유현저차이성( P<0.001),Lumican mRNA재대장암조직표체명현증고[이정상조직위1,암조직Lumican기인표체상조(5.35±0.66)배]。64례대장암조직Lumican단백양성표체솔위73.44%(47/64),현저고우원리종류적정상조직표체솔0%(0/27)(P<0.05)。Lumican표체여종류적병리분급、Dukes분기、림파결시부전이유관(P<0.001),이여환자성별、년령、병변부위무관(P>0.05)。27례대장암표본Lu-mican단백량여mRNA상대표체량정정상관(r=0.123),단무통계학의의(P>0.05)。결론:인대장암조직Lumican mRNA급단백균표체승고,Lumican고표체여대장암적림상병리특정급침습、전이특성밀절상관,재대장암발생、발전과정가능기착중요작용。Lumican가작위판단대장암생물학행위적중요지표지일。
Objective:To study the correlation between the expression levels of Lumican gene and protein in color-ectal carcinoma tissues and its clinicopathological significance. Methods:The mRNA levels of Lumican gene of 27 ca-ses of colorectal carcinoma and distant normal mucosa was quantified by RT-qPCR. The expression of Lumican was measured in 64 cases of colorectal carcinoma tissues and 27 cases of normal corresponding colorectal tissues by immu-nohistochemistry. Results:The expression of Lumican mRNA in colorectal carcinoma was obviously higher than that in distant normal mucosa(P<0. 001). The positive rate of Lumican protein expression in colorectal carcinoma and dis-tant normal mucosa was 73. 44% and 0%. The expressions of Lumican in colorectal carcinoma were significantly cor-related with the tumor differentiation,the Dukes stage and lymph node metastasis. There was no correlation with the expression of Lumican and the patientsˊsex,age and the position of the tumor. The expression of Lumican protein was positively correlated with Lumican mRNA in the same colorectal carcinoma,but this was not statistically significant( P>0. 05). Conclusion:The Lumican proteins and mRNA in colorectal carcinoma were significantly higher than those in distant normal mucosa. The expressions of Lumican in colorectal carcinoma were significantly correlated with the tumor differentiation,Dukes stage and lymph node metastasis. Lumican maybe played an important role in colorectal cancer.
