目的:探讨中药复骨方对糖皮质激素性骨质疏松(GIOP)大鼠骨折愈合的促进作用。方法将48只SD大鼠随机分为三组,各16只。前8周实验中,基础组仅给予肌内注射生理盐水1 mg/kg,模型组和治疗组肌内注射地塞米松1 mg/kg,1次/d,3次/周,建立GIOP动物模型。第8周末对三组大鼠手术造成单侧股骨闭合性骨折,骨折处采用金属内固定,建立GIOP骨折动物模型。术后第2天开始,基础组和模型组给予蒸馏水10 mL/kg灌胃,治疗组以复骨方药液灌胃给药,持续8周。观察各组股骨骨密度(BMD)变化,影像学观察骨折愈合情况,比较各组骨组织形态计量学参数和生物力学测量结果。结果治疗组股骨全长骨BMD高于模型组,差异有统计学意义(t=11.2532,P<0.05)。模型组大鼠股骨骨小梁面积百分数(TbAr豫)、骨小梁数量(TbN)、平均骨小梁厚度(TbTh)均较基础组下降,骨小梁分离度(TbSp)较基础组增高,差异有统计学意义(t=3.5885、7.4488、9.0512、10.0270,P<0.05);治疗组TbAr豫、TbN、TbTh均高于模型组,TbSp低于模型组,两组各指标比较差异有统计学意义(t=2.3329、5.3328、5.9512、10.9590,P<0.05)。模型组的最大载荷、最大挠度及最大应力均较基础组降低,差异有统计学意义(t =3.8944、5.3915、4.4739,P<0.05);治疗组的最大载荷、最大挠度及最大应力均高于模型组,差异有统计学意义(t =2.9251、3.7628、6.6926,P<0.05)。结论中药复骨方可提高GIOP骨折大鼠的BMD,改善骨微结构,提高骨生物力学性能。中药复骨方对GIOP骨折的愈合有促进作用。

목적:탐토중약복골방대당피질격소성골질소송(GIOP)대서골절유합적촉진작용。방법장48지SD대서수궤분위삼조,각16지。전8주실험중,기출조부급여기내주사생리염수1 mg/kg,모형조화치료조기내주사지새미송1 mg/kg,1차/d,3차/주,건립GIOP동물모형。제8주말대삼조대서수술조성단측고골폐합성골절,골절처채용금속내고정,건립GIOP골절동물모형。술후제2천개시,기출조화모형조급여증류수10 mL/kg관위,치료조이복골방약액관위급약,지속8주。관찰각조고골골밀도(BMD)변화,영상학관찰골절유합정황,비교각조골조직형태계량학삼수화생물역학측량결과。결과치료조고골전장골BMD고우모형조,차이유통계학의의(t=11.2532,P<0.05)。모형조대서고골골소량면적백분수(TbAr예)、골소량수량(TbN)、평균골소량후도(TbTh)균교기출조하강,골소량분리도(TbSp)교기출조증고,차이유통계학의의(t=3.5885、7.4488、9.0512、10.0270,P<0.05);치료조TbAr예、TbN、TbTh균고우모형조,TbSp저우모형조,량조각지표비교차이유통계학의의(t=2.3329、5.3328、5.9512、10.9590,P<0.05)。모형조적최대재하、최대뇨도급최대응력균교기출조강저,차이유통계학의의(t =3.8944、5.3915、4.4739,P<0.05);치료조적최대재하、최대뇨도급최대응력균고우모형조,차이유통계학의의(t =2.9251、3.7628、6.6926,P<0.05)。결론중약복골방가제고GIOP골절대서적BMD,개선골미결구,제고골생물역학성능。중약복골방대GIOP골절적유합유촉진작용。
Objective To observe the effects of Chinese Medicine Fugu Prescription on fracture healing of rats with glu-cocorticoid-induced osteoporosis (GIOP). Methods 48 SD rats were randomly divided into 3 groups, with 16 rats in each group. Rats of basic group were received intramuscular injection of 1 mg/kg saline. Rats of other groups given in-tramuscular injection of Dexamethasone (1 mg/kg, once a day, 3 times a week) for 8 weeks to establish GIOP animal model. In the end of 8th week, all rats were established the bone fracture in the middle of right femora. On the 2nd day of modeling, basic and model group were given distilled water 10 mL/kg by intragastric administration, treatment group was given Chinese Medicine Fugu Prescription, once per day for 8 weeks. Bone mineral density (BMD) was examined by dual-energy X-ray absorptiometry. Rats were sacrificed after fracture 8 weeks, femurs were removed for biomechan-ics test and morphologic observation. Results The BMD value of mice femora in the treatment group was higher than that in the model group, there was statistically significant difference between them (t = 11.2532, P< 0.05). The area percent of trabecular (TbAr%), the number and the thickness of trabecular (TbN, TbTh) in the model group were all lower than those in the basic group, but the trabecular bone space (TbSp) was higher than that in the basic group, there were statistically significant differences (t =3.5885, 7.4488, 9.0512, 10.0270, P< 0.05). The TbAr%, TbN, TbTh in the treatment group were higher than those in the model group, while TbSp was lower than that in the model group, there were statistically significant differences ( t =2 . 3329 , 5.3328, 5.9512, 10.9590, P<0.05). The maximum load, the maximum deflection and maximum stress in mod-el group were lower than those in the basic group, there were statistically significant differences (t = 3.8944, 5.3915, 4.4739, P < 0.05 ). The maximum load, the maximum deflection and maximum stress in the treatment group were higher than those in the model group, there were significant differences (t =2.9251, 3.7628, 6.6926, P<0.05). Con-clusion Chinese Medicine Fugu Prescription can increase BMD of GIOP rats with bone fracture, improve bone mi-crostructure, improve bone biomechanical properties. Chinese Medicine Fugu Prescription can promote fracture healing of rats with GIOP.
