目的:研究去氨加压素DDAVP调节豚鼠耳蜗功能及上皮钠通道蛋白α亚基(α-ENaC)的表达,探讨其在内淋巴积水中的作用。方法将24只豚鼠随机分为3组,分别给予0、5、10天DDAVP,通过耳蜗电图(ECochG)检测耳蜗功能学改变。应用苏木素-伊红染色(HE)确定各组耳蜗组织形态学改变,并用免疫组化法确定α-ENaC蛋白在耳蜗定位表达。采用蛋白印迹实验(WesternBlot)分别检测不同给药时间豚鼠耳蜗α-ENaC蛋白表达水平。结果DDAVP注射后,ECochG提示部分豚鼠耳蜗出现内淋巴积水,HE证实豚鼠内淋巴积水。α-ENaC蛋白在耳蜗血管纹、螺旋韧带、Reissner膜、Corti器、螺旋缘、螺旋神经、耳蜗神经表达。与未给予DDAVP相比,10天组豚鼠耳蜗α-ENaC蛋白表达升高(P<0.05);。结论 DDAVP调节豚鼠耳蜗α-ENaC蛋白表达升高,进而调节内淋巴代谢,诱导内淋巴积水,影响内耳功能。

Objective To investigate effects of desmopressin (DDAVP) on epithelial sodium channel (ENaC) expres-sion in the cochlea, and to study its possible role in the etiology of endolymphatic hydrops. Methods Twenty-four guinea pigs were divided into 3 groups. They received systemic injection of desmopressin for 0, 5 or 10 days respectively. The cochle-ae were examined by electrocochleography (ECochG), stained with HE and immunohistochemistry, and tested with Western blot. Results After being treated with DDAVP, ECochG showed mild endolymphatic hydrops in one guinea pig which was ver-ified by HE staining.α-ENaC was expressed in the stria vascularis, spiral ligament, Reissner’s membrane, organ of Corti, spiral limbus, spiral ganglions and cochlear nerve. Western blot showed that α-ENaC protein in the cochlea increased in DDAVP-treated guinea pigs (P<0.05). Conclusion Desmopressin increases the expression of α-ENaC in the cochlea, which may induce endolymphatic hydrops and affect function of the inner ear.
