目的:探讨肝细胞生长因子( HGF)对人脐带间充质干细胞( hUCMSCs)增殖、黏附及迁移能力的影响。方法按照不同腺病毒感染 hUCMSCs 分为实验组( Ad-GFP-HGF )和对照组( Ad-GFP )。以最佳感染复数转染hUCMSCs,观察细胞形态变化以及GFP蛋白的表达。比较各组细胞增殖能力、黏附能力及迁移能力。 Western Bolt检测各组细胞HGF的表达水平。结果对照组和实验组均出现了GFP的表达,实验组hUCMSCs的增殖、黏附及迁移能力均较对照组增强(P<0.05)。实验组细胞HGF的表达水平显著高于对照组(P<0.05)。结论 HGF的高表达能明显促进hUCMSCs的增殖、黏附和迁移。

목적:탐토간세포생장인자( HGF)대인제대간충질간세포( hUCMSCs)증식、점부급천이능력적영향。방법안조불동선병독감염 hUCMSCs 분위실험조( Ad-GFP-HGF )화대조조( Ad-GFP )。이최가감염복수전염hUCMSCs,관찰세포형태변화이급GFP단백적표체。비교각조세포증식능력、점부능력급천이능력。 Western Bolt검측각조세포HGF적표체수평。결과대조조화실험조균출현료GFP적표체,실험조hUCMSCs적증식、점부급천이능력균교대조조증강(P<0.05)。실험조세포HGF적표체수평현저고우대조조(P<0.05)。결론 HGF적고표체능명현촉진hUCMSCs적증식、점부화천이。
Objective To explore the effect of hepatocyte growth factor ( HGF) on the proliferation, adhesion and migration of human umbilical cord mesenchymal stem cells ( hUCMSCs) . Methods The hUCMSCs were divided in-to experimental group ( Ad-GFP-HGF) and control group ( Ad-GFP) according to the different adenovirus infection. hUCMSCs was transfected by the optimal MOI ( multiplicity of infection) to observe the changes in cell morphology and the GFP (green fluorescent protein) expression. The proliferation, adhesion and migration abilities of cells in the two groups were compared. The HGF expression level in each group was detected by Western Bolt method. Results The positive GFP expression was found in the experimental and control groups, and the proliferation, adhesion and migration abilities of cells in experimental group significantly improved, compared with those in control group ( P<0. 05 ) . The HGF expression level in experimental group was significantly higher than that in control group (P<0. 05). Conclusion The high expression of HGF can significantly promote the proliferation, adhesion and migration of hUCMSCs.
