目的:观察高果糖饮食喂养对大鼠骨骼肌内长链酯酰辅酶A( LCACoAs)含量的影响,并探讨高果糖饮食喂养大鼠肌细胞内LCACoAs与胰岛素抵抗( IR)的关系。方法 Wistar雄性大鼠40只随机分为对照组及高果糖组,每组20只。对照组进食普通饲料,高果糖组饮食中果糖占总热量34.5%。喂养8周,测定各组大鼠空腹血糖( FBG)、空腹胰岛素( FINS)、肌肉甘油三酯( TG)及肌细胞内LCACoAs的含量,正常葡萄糖高胰岛素钳夹技术测定葡萄糖输注率(GIR),并用RT-PCR法测定脂酰辅酶A合成酶1(ACS1)的基因表达。结果与对照组相比,高果糖组的FBG和FINS浓度、肌内TG和LCACoAs含量、ACS1的基因表达均明显增高,而GIR明显降低( P<0.01);TG和LCA-CoAs含量与GIR均呈负相关(P<0.01)。结论高果糖饮食可引起机体IR及肌肉LCACoA的堆积,肌肉LCACoA增多与ACS基因表达增高有关。

목적:관찰고과당음식위양대대서골격기내장련지선보매A( LCACoAs)함량적영향,병탐토고과당음식위양대서기세포내LCACoAs여이도소저항( IR)적관계。방법 Wistar웅성대서40지수궤분위대조조급고과당조,매조20지。대조조진식보통사료,고과당조음식중과당점총열량34.5%。위양8주,측정각조대서공복혈당( FBG)、공복이도소( FINS)、기육감유삼지( TG)급기세포내LCACoAs적함량,정상포도당고이도소겸협기술측정포도당수주솔(GIR),병용RT-PCR법측정지선보매A합성매1(ACS1)적기인표체。결과여대조조상비,고과당조적FBG화FINS농도、기내TG화LCACoAs함량、ACS1적기인표체균명현증고,이GIR명현강저( P<0.01);TG화LCA-CoAs함량여GIR균정부상관(P<0.01)。결론고과당음식가인기궤체IR급기육LCACoA적퇴적,기육LCACoA증다여ACS기인표체증고유관。
Objective To observe the effect of high-fructose feeding on the content of long chain acyl-CoA ( LCACoA) in skeletal muscles in rats, and to study the relationship of LCACoAs and insulin resistance ( IR) in muscle cells fed with high-fructose in rats. Methods Forty male Wistar rats were randomly divided into control group and high-fructose group ( n=20 for each group) . Control group was fed with common diet, while high-fructose group was fed with 34. 5% total calories of high-fructose diet. After 8 weeks of feeding, levels of fasting blood glucose (FBG), fasting serum insulin ( FINS) , contents of muscle triglyceride ( TG) and LCACoAs in muscle tissues were measured. Hyperinsuline-mic-euglycemic clamp study was performed to evaluate whole-body insulin sensitivity ( glucose infusion rate, GIR) , and the gene expression of fatty acyl-coa synthetase 1 ( ACS1 ) was detected by RT-PCR method. Results Compared with those in control group, the levels of FBG, FINS, contents of muscle TG and total LCACoA in muscle tissues and gene ex-pression of ACS1 were significantly increased, while GIR level was significantly decreased in high-fructose group; con-tents of muscle TG and total LCACoA in muscles was negatively correlated with GIR (P<0. 01). Conclusion Whole-body insulin resistance and LCACoAs accumulation can be induced by high-fructose feeding, and the increase of LCACoA content in muscle might be associated with the increased gene expression of ACS.
