目的:探讨短期能量限制( CR)对正常大鼠胰岛素敏感性的影响及其机制。方法将24只F344/NSIc系雄性大鼠随机分为对照组( AL组)和能量限制组( CR组)各12只。 AL组自由摄入食物及水,CR组限制食物摄入(不禁水)8周(饲料总摄入量为对照组的64%)。采用高胰岛素-正常血糖钳夹试验测定两组葡萄糖输注率( GIR)判断其胰岛素敏感性( GIR越高,胰岛素敏感性越高);比较两组体质量增加及内脏脂肪重量;Western blot法检测骨骼肌葡萄糖转运体4(GluT4)、蛋白激酶B底物蛋白160(AS160)及磷酸化AS160(p-AS160)蛋白表达。结果 CR组GIR明显高于对照组,体质量及内脏脂肪重量明显低于对照组;两组GluT4、AS160及p-AS160蛋白表达无显著差异。结论短期CR可提高大鼠正常状态下的胰岛素敏感性,其机制可能与AS160上游的胰岛素信号转导蛋白有关。

목적:탐토단기능량한제( CR)대정상대서이도소민감성적영향급기궤제。방법장24지F344/NSIc계웅성대서수궤분위대조조( AL조)화능량한제조( CR조)각12지。 AL조자유섭입식물급수,CR조한제식물섭입(불금수)8주(사료총섭입량위대조조적64%)。채용고이도소-정상혈당겸협시험측정량조포도당수주솔( GIR)판단기이도소민감성( GIR월고,이도소민감성월고);비교량조체질량증가급내장지방중량;Western blot법검측골격기포도당전운체4(GluT4)、단백격매B저물단백160(AS160)급린산화AS160(p-AS160)단백표체。결과 CR조GIR명현고우대조조,체질량급내장지방중량명현저우대조조;량조GluT4、AS160급p-AS160단백표체무현저차이。결론단기CR가제고대서정상상태하적이도소민감성,기궤제가능여AS160상유적이도소신호전도단백유관。
Objective To investigate the effect of the short-term caloric restriction( CR) on insulin sensitivity and its signal transduction mechanisms.Methods A total of 24 F344/NSIc rats were equally divided into ad libitum group ( AL group) and CR group.Rats in AL group were free fed every alternate day for 8 weeks, while rats in CR group were fed re-strict caloric without water deprivation for 8 weeks.Glucose infusion rate(GIR) and insulin sensitivity were measured by u-sing hyperinsulinemic-euglycemic clamp method.The body weight mass and visceral fat weight between the two groups were compared.The muscle glucose transporter 4(GluT4), kinase substrate 160(AS160 ) protein and p-AS160 protein was de-tected by using Western blot method.Results GIR in CR group was obviously higher than that of control group, while body mass and visceral fat weight significantly lower than control group.However, there was no significant difference in ex-pression of GluT4, AS160 protein and p-AS160 protein.Conclusions Short-term caloric restriction can improve the insu-lin sensitivity in healthy rats which may be associated with reduced the visceral fat increasing.
