利用ISSR分子标记技术对云南省选育出的49个板栗优良品种进行遗传多样性和亲缘关系分析。结果表明,筛选出的11条引物一共扩增出86条DNA谱带,其中70条为多态性条带,多态位点百分率( PPB %)81.40%,多态性指数(H)0.228, Shannon信息指数(I)0.350,数据表明所选育的板栗优良品种资源具有较高的遗传多样性。根据各品种的Nei’ s遗传距离,进行UPGMA聚类,49个品种被分为2个大类,其中第1大类又分为了2个亚类,来源地相同的品种基本聚在同一类。聚类结果在分子水平上揭示了各板栗品种的亲缘关系,为今后板栗育种的亲本选配提供科学依据。

Inter-simple Sequence Repeat ( ISSR) molecular markers were used to analyze genetic diversity and re-lations of 49 chestnut cultivars (Castanea mollissima).86 bands were amplified by 11 selected primers, among which 70 bands were polymorphic .Percentage of polymorphic loci was 81.4%, the mean expected heterozygosity (H) was 0.228 and the Shannon information index (I) was 0.350.The results indicated that selected chestnut cultivars had a high level of genetic diversity .According to Nei ’ s genetic distances , 49 cultivars were clustered into 2 groups and 4 subgroups by the UPGMA analysis . The clustering accorded mainly with their introduced places.The genetic relationship of all 49 chestnut cultivars revealed by the ISSR dendrogram would be help to ac-curately select the parents in chestnut breeding programs .
