为了研究泛素连接酶 Nedd4对肿瘤细胞增殖和凋亡的影响，利用小 RNA 干扰技术，通过慢病毒包装，侵染前列腺癌细胞 DU145，达到下调 Nedd4表达的目的．MTT 检测表明，下调 Nedd4表达可以抑制 DU145细胞的增殖；DAPI 染色发现，下调 Nedd4表达的 DU145细胞对星形孢菌素诱导的细胞凋亡更加敏感．这些结果都说明，下调Nedd4的表达不仅可以抑制DU145肿瘤细胞增殖，而且可以促进细胞凋亡．

To study the effects of Nedd4 on tumor cell proliferation and apoptosis,Nedd4 was depleted in prostate cancer cell DU145 by infecting the lentiviruses expression of Nedd4-shRNAs. MTT assay showed that that depletion of Nedd4 de-creased DU145 cell proliferation. DAPI staining indicated that knockdown of Nedd4 resulted in greater sensitivity to stauro-sporine-induced apoptosis of DU145 cells. These results revealed that the down-regulation of Nedd4 expression not only reduced the cell proliferation but also induced the apoptosis of DU145 cells.
