中药五陪子能够有效预防和治疗龋齿,但其抗龋活性成分和机制尚待研究.本实验应用不同剂量五倍子大极性非酚性成分处理根面龋大鼠模型,Bowen记分法评价实验动物根面龋情况, ELISA法检测大鼠龈沟液内基质金属蛋白酶-9(MMP-9)的表达变化.结果发现1 mg/mL及2 mg/mL五倍子处理组第一磨牙根面龋记分及龈沟液内MMP-9含量均显著低于蒸馏水处理组和0.5 mg/mL五倍子处理组(P<0.01),与氟化钠处理组间无显著性差异(P>0.05),且两组间比较无显著性差异(P>0.05).说明五倍子大极性非酚性物质能够通过抑制龋齿饮食大鼠龈沟液内MMP-9表达以抑制根面龋进展,最佳浓度为1 mg/mL.

Galla Chinensis plays an effective role in preventing and treating caries.However,the active compo-nent and its mechanism still remain unknown.Different doses of non-phenolic constituents of Galla Chinensis extracts(GCE)are used to treat root surface caries in SD rats.Root surface caries is estimated by a modified Bowen′s method,and the levels of matrix metalloproteinase-9(MMP-9)in gingival crevicular fluid are detec-ted by ELISA.The results indicate that Bowen′s scores and the levels of MMP-9 in moderate or high dose GCE group are much less than those in control group or low dose GCE group(P<0.01).These above indexes are sim-ilar to those in NaF group(P>0.05 ).There is no difference between moderate and high dose GCE groups(P>0.05 ).It is therefore concluded that the non-phenolic constituents of GCE can prevent the progress of root sur-face caries by inhibiting the expression of MMP-9 in rats.The optimum working concentration is 1 mg/mL.
