目的:通过研究变应性鼻炎大鼠鼻黏膜 NF-κBp65、GATA-3表达的影响,探讨玉屏风散治疗变应性鼻炎的可能机制。方法60只SD 大鼠按体质量随机分为6组,每组10只,分别为正常组、模型组、治疗组(玉屏风散低、中、高剂量组)、对照组。除正常组外,其余的大鼠均建立卵清蛋白诱导的变应性鼻炎模型。分组给药干预1周。应用 HE 染色检测鼻黏膜组织结构变化,应用 western-blot 及 Real-time PCR 技术检测 NF-κBp65、GATA-3在鼻黏膜组织中的表达。结果模型组鼻组织病理学表现以组织重塑、上皮细胞脱落、杯状细胞化生、组织水肿、大量嗜酸性粒细胞浸润为主;治疗组的表现以鼻黏膜存在组织重塑表现,鼻黏膜上皮细胞脱落、组织间质轻度水肿、固有层可见少量嗜酸性粒细胞浸润,两组比较,有统计学差异(P <0.05)。模型组鼻黏膜中 NF-κBp65、GATA-3蛋白及其 mRNA 呈高表达变化;给予玉屏风散治疗后,NF-κBp65、GATA-3蛋白及其 mRNA 表达下降,经统计学比较,有显著性差异(P <0.05)。结论变应性鼻炎鼻黏膜存在组织重塑,玉屏风散可能通过下调 NF-κBp65、GATA-3表达,发挥防治变应性鼻炎的效应。

목적:통과연구변응성비염대서비점막 NF-κBp65、GATA-3표체적영향,탐토옥병풍산치료변응성비염적가능궤제。방법60지SD 대서안체질량수궤분위6조,매조10지,분별위정상조、모형조、치료조(옥병풍산저、중、고제량조)、대조조。제정상조외,기여적대서균건립란청단백유도적변응성비염모형。분조급약간예1주。응용 HE 염색검측비점막조직결구변화,응용 western-blot 급 Real-time PCR 기술검측 NF-κBp65、GATA-3재비점막조직중적표체。결과모형조비조직병이학표현이조직중소、상피세포탈락、배상세포화생、조직수종、대량기산성립세포침윤위주;치료조적표현이비점막존재조직중소표현,비점막상피세포탈락、조직간질경도수종、고유층가견소량기산성립세포침윤,량조비교,유통계학차이(P <0.05)。모형조비점막중 NF-κBp65、GATA-3단백급기 mRNA 정고표체변화;급여옥병풍산치료후,NF-κBp65、GATA-3단백급기 mRNA 표체하강,경통계학비교,유현저성차이(P <0.05)。결론변응성비염비점막존재조직중소,옥병풍산가능통과하조 NF-κBp65、GATA-3표체,발휘방치변응성비염적효응。
Objestive To study the changes of NF-κBp65 and GATA-3 in allergic rhinitis. The mechanism of Yupingfeng decoction interfered with AR was discussed. Method Sixty SD rats were divided into six groups by weight. Such as normal group, model group, treatment group (Yupingfengsan low, medium and high dose group), the control group. Except normal group, the AR models are established by OVA-induced. The histopathology changes were detected by HE stains. The expression of NF-κBp65 and GATA-3 was assayed by Real-time PCR and Western-blot. Result The AR in blank group is characterized by tissue remodel and epithelial cells abscission as well as Goblet cells metaplasia and eosinophil infiltration in the tissue. However, the histopathology changes in intervention group are characterized by tissue remodel and dermal edema and infiltration of little eosinophil cells. There is an obvious different between the two groups(P<0.05). The expressions of NF-κBp65 and GATA-3 are in the increment in blank group. But the expressions of NF-κBp65 and GATA-3 are on the decline in intervention(P<0.05). Conclusion Tissue remodeling exists in nasal mucosa of the allergic rhinitis rat models. Yupingfeng decoction exerts a distinct effect on preventing the onset of AR by down-regulating the expression of NF-κBp65 and GATA-3.
