目的:研究不同浓度的大豆异黄酮对体外培养大鼠成骨细胞增殖活性和骨钙素(BGP)基因表达的影响。方法采用新生大鼠颅骨分离培养成骨细胞,加入不同浓度的大豆异黄酮(20、40、60、80μg/μL),另设空白对照组。MTT法测成骨细胞增殖活性。提取细胞总RNA,RT-PCR半定量分析细胞BGPmRNA表达。结果MTT结果显示,60和80μg/μL组成骨细胞增殖活性与对照组相比显示增高,有统计学意义(P<0.05)。RT-PCR结果显示:60和80μg/μL组BGPmRNA表达增强,有统计学意义(P<0.01)。结论大豆异黄酮可以促进成骨细胞的增殖活性和BGP基因上调,并呈明显的剂量依赖关系。

목적:연구불동농도적대두이황동대체외배양대서성골세포증식활성화골개소(BGP)기인표체적영향。방법채용신생대서로골분리배양성골세포,가입불동농도적대두이황동(20、40、60、80μg/μL),령설공백대조조。MTT법측성골세포증식활성。제취세포총RNA,RT-PCR반정량분석세포BGPmRNA표체。결과MTT결과현시,60화80μg/μL조성골세포증식활성여대조조상비현시증고,유통계학의의(P<0.05)。RT-PCR결과현시:60화80μg/μL조BGPmRNA표체증강,유통계학의의(P<0.01)。결론대두이황동가이촉진성골세포적증식활성화BGP기인상조,병정명현적제량의뢰관계。
Objective To study the effect of different concentrations of isoflavones on gene expression of BGP mRNA and cell proliferation activity in cultured rat osteoblasts. Methods Newborn rat calvarid osteoblastic cell were isolated and cultured. They were cultured in medium with various concentration isoflavones(20, 40, 60, 80 μg/μL), compared to control group. The proliferation activity by MTT method. The total RNA was extracted by TRIZOL and analyaed with semi-quantitative RT-PCR. Results Compared to control group, the cell proliferation activity of 60 and 80 μg/μLgroup was significantly promoted(P<0.05), the cell BGP mRNA expression of 60 and 80 μg/μL group was significantly upregulated(P<0.01). Conclusion The isofiavones could increase cell proliferation activity and expression of BGP mRNA. The effect was similar to estrogen.
