Common turmeric%N-butyl alcohol%Pancreas fibrosis%MASSON staining
目的:观察温郁金正丁醇提取物对二乙基二硫代氨基甲酸钠(DDC)诱导的大鼠胰腺纤维化的影响。方法66只雄性、健康、清洁级SD大鼠,按随机数字表法分为空白对照组(A组,n=12)、模型组(B组,n=18)、温郁金正丁醇提取物高剂量组(C组,n=18)、温郁金正丁醇提取物低剂量组(D组,n=18)。经腹腔注射DDC溶液制备大鼠CP胰腺纤维化模型(除空白对照组外),空白对照组大鼠在正常饲养1周后,每天予以生理盐水灌胃(每次1ml/kg,1次/d);模型组在造模1周后,造模同时,每天予以生理盐水灌胃(每次1ml/kg,1次/d);温郁金正丁醇提取物高剂量组造模1周后,在造模同时,每天予以高剂量温郁金正丁醇提取物灌胃(每次剂量相当于温郁金生药5g/kg,1次/d);温郁金正丁醇提取物低剂量组造模1周后,在造模同时,每天予以低剂量温郁金正丁醇提取物灌胃(每次剂量相当于温郁金生药1.25g/kg,1次/d)。第10周,麻醉大鼠,取胰腺组织,行MASSON染色,测定胰腺胶原纤维面积。结果空白对照组胰腺组织中,在血管周围可见少量胶原纤维;模型组大鼠胰腺中胶原纤维广泛分布于胰腺小叶内或小叶间,较密集,胶原纤维面积百分比为(13.46±5.58)%;温郁金正丁醇提取物高、低剂量干预组(C、D组)胰腺胶原纤维面积百分比为(3.34±1.48)%、(3.51±2.19)%,干预组胰腺胶原纤维比模型组均明显减少(P均<0.05)。干预组间比较,胰腺胶原纤维面积差异无统计学意义。结论温郁金正丁醇提取物能缓解DDC诱导的慢性胰腺炎(CP)大鼠胰腺纤维化进程。

목적:관찰온욱금정정순제취물대이을기이류대안기갑산납(DDC)유도적대서이선섬유화적영향。방법66지웅성、건강、청길급SD대서,안수궤수자표법분위공백대조조(A조,n=12)、모형조(B조,n=18)、온욱금정정순제취물고제량조(C조,n=18)、온욱금정정순제취물저제량조(D조,n=18)。경복강주사DDC용액제비대서CP이선섬유화모형(제공백대조조외),공백대조조대서재정상사양1주후,매천여이생리염수관위(매차1ml/kg,1차/d);모형조재조모1주후,조모동시,매천여이생리염수관위(매차1ml/kg,1차/d);온욱금정정순제취물고제량조조모1주후,재조모동시,매천여이고제량온욱금정정순제취물관위(매차제량상당우온욱금생약5g/kg,1차/d);온욱금정정순제취물저제량조조모1주후,재조모동시,매천여이저제량온욱금정정순제취물관위(매차제량상당우온욱금생약1.25g/kg,1차/d)。제10주,마취대서,취이선조직,행MASSON염색,측정이선효원섬유면적。결과공백대조조이선조직중,재혈관주위가견소량효원섬유;모형조대서이선중효원섬유엄범분포우이선소협내혹소협간,교밀집,효원섬유면적백분비위(13.46±5.58)%;온욱금정정순제취물고、저제량간예조(C、D조)이선효원섬유면적백분비위(3.34±1.48)%、(3.51±2.19)%,간예조이선효원섬유비모형조균명현감소(P균<0.05)。간예조간비교,이선효원섬유면적차이무통계학의의。결론온욱금정정순제취물능완해DDC유도적만성이선염(CP)대서이선섬유화진정。
Objective To observe the effect of common turmeric n-butanol extraction on the pancreas fibrosis in chronic pancreatitis rat induced by Diethyldithiocarbamic acid sodium salt(DDC). Methods 66 male,health and clean Sprague-Dawley rats,divided randomly into the group of blank control(group A,n=12),the group of model(group B,n=18),the group of high dose common turmeric extraction by n-butanol(group C,n=18),the group of low dose common turmeric extraction by n-butanol(group D,n=18). The rats model of pancreas fibrosis were made with intraperitoneal injection DDC except the group of blank control. After mading model one week,the rats of group A were given normal saline(1ml/kg)by gavage after one week normally fed;the rats of group B given normal saline(1ml/kg)by gavage every day,the rats of group C given high dose common turmeric n-butanol-extraction(equivalent to the crude herb 5g/kg)by gavage every day,the rats of group D given low dose common turmeric n-butanol-extraction(equivalent to the crude herb 1.25g/kg)by gavage every day,at the same time,the rats were mading model. After ten weeks,anaesthetized the rats,got pancreas tissues,measured the aera of collagen fiber in MASSON staining. Results In control group,the distribution of collagen fiber were aroud the vascular.In model group,the collagen fiber was widely and densely distributed between the pancreatic lobules,and the percentage of collagen fiber in model group was(13.46±5.58)%,the percentage of collagen fiber in high dose common turmeric n-butanol-extraction group was(3.34±1.48)%. The percentage of collagen fiber in low dose common turmeric n-butanol-extraction group was (3.51±2.19)%,the collagen fiber in intervention groupgroup was much less than model group for all(P<0.05 for all).Compared with high dose common turmeric n-butanol-extraction group,the collagen fiber area of pancreas in low dose common turmeric n-butanol-extraction group was similar,and the difference was not significant(P>0.05). Conclusion The extraction by n-butyl alcohol in common turmeric can reduce the collagen fiber in pancreas fibrosis of chronic pancreatitis rat induced by DDC.
