目的:探讨c-Met基因在胶质中的表达及沉默c-Met对脑胶质瘤细胞生物学行为的影响。方法选择良好生长的U87胶质瘤细胞,在培养70%左右密度时进行传代。在转染中,对照组转染空载体PGC,实验组转染PGC-c-Met shRNA载体,然后进行细胞增殖、细胞周期与细胞凋亡的检测。结果两组均得到稳定转染的U87胶质瘤细胞,实验组的细胞生长计数明显少于对照组(P<0.05)。经过流式细胞仪检测,相对于对照组,实验组大部分细胞处于G0期,S期细胞明显减少,差异均有统计学意义(P<0.05)。同时实验组的U87细胞凋亡明显增加(P<0.05)。结论 HGF及c-Met在脑胶质瘤中表达广泛,沉默c-Met能阻止U87胶质瘤细胞的生长,其作用的发挥与调控胶质瘤细胞的细胞周期与细胞凋亡有关。

목적:탐토c-Met기인재효질중적표체급침묵c-Met대뇌효질류세포생물학행위적영향。방법선택량호생장적U87효질류세포,재배양70%좌우밀도시진행전대。재전염중,대조조전염공재체PGC,실험조전염PGC-c-Met shRNA재체,연후진행세포증식、세포주기여세포조망적검측。결과량조균득도은정전염적U87효질류세포,실험조적세포생장계수명현소우대조조(P<0.05)。경과류식세포의검측,상대우대조조,실험조대부분세포처우G0기,S기세포명현감소,차이균유통계학의의(P<0.05)。동시실험조적U87세포조망명현증가(P<0.05)。결론 HGF급c-Met재뇌효질류중표체엄범,침묵c-Met능조지U87효질류세포적생장,기작용적발휘여조공효질류세포적세포주기여세포조망유관。
Objective To investigate the human glioma cells biological behavior effects by the gene silencing of c-Met expression. Methods Selected better growth U87 glioma cells,when it were cultured density of about 70%were passaged . In the transfection, the control group was transfected with empty vector PGC,the experimental group was transfected with PGC-c-Met shRNA vectors,then they were given the cell proliferation,cell cycle and cell investigation. Results The two groups had been stably transfected U87 glioma cells. The cell growth counts in the experimental group were significantly less than the control group(P<0.05). After flow cytometry, 0uuyr45t’kj76776rdel;ui,89meta. Conclusion HGF and c-met were widely expressioned,silencing the expression of c-met can significantly prevent the growth of U87 glioma cells ,its roles may be related to cell cycle regulation and cells apoptosis.
