为探索小麦简便、快速的活力检测方法,采用刃天青-酵母混合试剂对单粒小麦种子进行浸泡,通过浸泡液颜色的变化判断种子活力高低。对小麦种子进行人工老化3d、6d以获得活力不同的种子,将人工老化后的3种不同活力种子平均分布于96孔板中,在35℃条件下反应后进行颜色观察,并用酶标仪在570 nm下进行吸光度检测。结果显示:160μL是适宜的反应体积,5.5 h是适宜的保温时间。相关分析表明,小麦种子活力、浸泡后种子的发芽率与5.5h刃天青浸泡液的吸光度值呈显著正相关,且蓝色部分的百分比与种子活力具有高度相关性。可以采用刃天青试剂颜色变化或570 nm平均吸光度值来快速、准确地预测小麦种子活力。

The seed vigor of wheat is important for food production ,to explore a simple and rapid test method for wheat seed vigor measurement with single seed using resazurin reagent(RR) which was made by resazurin and yeast mixture ,through RR liquid color changed from blue to pink or colorless to reflect the seed vigor . The testing theory is matter leaching from seed membrane containing organic compounds such as glucose and ethanol ,which can make RR changed from blue to pink or colorless ,the more seed membrane leaked ,the more RR changed in unit time .Seeds were artificially aged 3 d ,6 d to create three contrasting levels of viabilities .Seeds with different vigor were evenly distributed and soaked into 96-well plate under the condition of 35 ℃ ,and absorbance of the RR was measured at 570 nm with a microplate reader ,then blue colors rate was counted .The results show that the suitable volume is 160 μL ,and the suitable reaction time is 5 .5 h . Through correlation analysis ,wheat seed vigor ,and germination percentage of the seeds after soaked were significantly correlated with their blue color fraction ,and average absorbance under 570nm after 5 .5 h soaked in RR .Therefore ,color changed in RR and average absorbance under 570 nm can be used to quickly and accurately predict wheat seed vigor .
