为筛选薏苡种仁内生真菌的抗菌活性,采用组织分离法,从禾本科黍亚科蜀黍族薏苡种仁中分离内生真菌,采用菌饼法筛选其抗菌活性。通过5.8 S ITS 分子系统学鉴定菌种,高效液相色谱法(HPLC)分析鉴定菌种发酵液正丁醇提取物次的生代谢产物。结果表明:分离共获得23株薏苡种仁内生真菌,其中,有弯孢霉属、链格孢属、砖孢属、茎点霉属、灰软盘菌属、稻黑孢属、玉蜀黍赤霉属、曲霉属及其他真菌。57%的菌株至少对测试菌株中的1种具有抗菌活性,且 GZR19号菌株具有广谱抗菌活性并鉴定为1种茎点霉(Phoma sp .),其发酵液的次生代谢产物在该色谱条件下主要有4种物质含量较高,其中可能存在具有抗菌活性的化合物。

The antimicrobial activity of endophytic fungi isolated from Coix lacryma-jobi kernels was screened by the colony method,then the strains were identified by 5.8 S ITS molecular systematics,and finally secondary metabolites extracted from strain fermentation liquor were analyzed by HPLC.The results showed that 23 endophytic fungi strains including Curvularia, Alternaria, Dictyo-mitosporicfungi ,Phoma sp .,Mollisia cinerea ,Nigrospora oryzae ,Gibberella zeae ,Aspergillus and other fungi are isolated.57% strains has the antimicrobial activity against one test strain at least.GZR19 strain with broad-spectrum antimicrobial activity is identified as Phoma sp.and there are main four substances in secondary metabolites of the fermentation liquor,which indicates that the compounds with antimicrobial activity may exist in four substances.
