目的:观察脓毒症大鼠肝脏组织中免疫相关基因表达的改变。方法将120只Wistar大鼠按随机数字表法分为假手术组30只、模型组90只,模型组根据选模后不同时间点(24、48、72 h)随机分为3个亚组各30只。通过盲肠结扎穿孔术( CLP)复制脓毒症大鼠模型,应用含有28000个大鼠基因cDNA克隆的基因芯片,检测脓毒症大鼠肝组织在CLP后各个时点的免疫相关基因表达变化并筛选出差异基因。结果造模24 h,筛选出差异基因671条,免疫相关基因97条(表达上调48条、下调49条);造模48 h,筛选出差异基因133条,免疫相关基因36条(表达上调13条、下调23条);造模72 h,筛选出差异基因204条,免疫相关基因33条(表达上调18条、下调15条)。3个时间点重复基因共58条,免疫相关基因15条(表达上调4条、下调10条、先上调后下调1条)。结论脓毒症大鼠肝组织中,一系列与细胞防御及免疫功能相关的基因表达出现显著改变,它们可能在不同程度上参与了脓毒症大鼠肝损伤的病理生理过程。
目的:觀察膿毒癥大鼠肝髒組織中免疫相關基因錶達的改變。方法將120隻Wistar大鼠按隨機數字錶法分為假手術組30隻、模型組90隻,模型組根據選模後不同時間點(24、48、72 h)隨機分為3箇亞組各30隻。通過盲腸結扎穿孔術( CLP)複製膿毒癥大鼠模型,應用含有28000箇大鼠基因cDNA剋隆的基因芯片,檢測膿毒癥大鼠肝組織在CLP後各箇時點的免疫相關基因錶達變化併篩選齣差異基因。結果造模24 h,篩選齣差異基因671條,免疫相關基因97條(錶達上調48條、下調49條);造模48 h,篩選齣差異基因133條,免疫相關基因36條(錶達上調13條、下調23條);造模72 h,篩選齣差異基因204條,免疫相關基因33條(錶達上調18條、下調15條)。3箇時間點重複基因共58條,免疫相關基因15條(錶達上調4條、下調10條、先上調後下調1條)。結論膿毒癥大鼠肝組織中,一繫列與細胞防禦及免疫功能相關的基因錶達齣現顯著改變,它們可能在不同程度上參與瞭膿毒癥大鼠肝損傷的病理生理過程。
목적:관찰농독증대서간장조직중면역상관기인표체적개변。방법장120지Wistar대서안수궤수자표법분위가수술조30지、모형조90지,모형조근거선모후불동시간점(24、48、72 h)수궤분위3개아조각30지。통과맹장결찰천공술( CLP)복제농독증대서모형,응용함유28000개대서기인cDNA극륭적기인심편,검측농독증대서간조직재CLP후각개시점적면역상관기인표체변화병사선출차이기인。결과조모24 h,사선출차이기인671조,면역상관기인97조(표체상조48조、하조49조);조모48 h,사선출차이기인133조,면역상관기인36조(표체상조13조、하조23조);조모72 h,사선출차이기인204조,면역상관기인33조(표체상조18조、하조15조)。3개시간점중복기인공58조,면역상관기인15조(표체상조4조、하조10조、선상조후하조1조)。결론농독증대서간조직중,일계렬여세포방어급면역공능상관적기인표체출현현저개변,타문가능재불동정도상삼여료농독증대서간손상적병리생리과정。
Objective To observe the change of immune related gene-expression profiling in rats with sepsis.Meth-ods 120 male wistar rats were randomly divided into sham operation group ( n=30 ) and model group ( n=90 ) .The model group according to different time points (24 h, 48 h and 72 h) were randomly divided into 3 groups ( each subgroup n=30).The rats of model group received cecal ligature and puncture (CLP) operation.Gene chips containing 28 000 rot-genes cDNA were used to test gene-expression in liver of septic rats to select the genes with different expressions with soft-ware.Results Model group had 671 differential expression genes with 97 immune related genes at 24 h;133 differential expression genes with 36 immune related genes at 48 h;204 differential expression genes with 33 immune related genes at 72 h.Three time points in model group and sham operation group repeated gene were 58, immune related gene 15.Con-clusion A series of cell defense and immune function genes change significantly in liver tissue of septic rats, which may be involved in the pathophysiology of sepsis induced hepatic injury in different extent.
