目的：探讨结肠癌组织中SLP-2基因的表达情况，及 SLP-2蛋白对结肠癌 HCT-116细胞增殖和细胞周期的影响。方法采用免疫组织化学法检测50例结肠癌及对应癌旁正常结肠组织中 SLP-2蛋白的表达，分析结肠癌病例临床病理特征与SLP-2蛋白表达的关系，将 SLP-2小干扰 RNA（siRNA）转染 HCT-116细胞，采用逆转录（RT）-PCR及 Western blot检测转染SLP-2 siRNA后 HCT-116细胞中基因及蛋白的表达情况，MTT法和流式细胞术分别检测转染 SLP-2 siRNA后 HCT-116细胞生长状况及细胞周期。结果癌旁正常结肠组织及结肠癌组织中 SLP-2的高表达率分别为22％和70％，差异有统计学意义（P＜0．05）；SLP-2表达情况与结肠癌分期及淋巴结转移情况相关（P＜0．05）。转染 SLP-2 siRNA后，HCT-116细胞中 SLP-2 mRNA和蛋白的表达均下降（P＜0．05），HCT-116细胞 G1期增高，S期减少，细胞生长变慢（P＜0．05）。结论 SLP-2在结肠癌中高表达，并参与结肠癌的发生发展。

목적：탐토결장암조직중SLP-2기인적표체정황，급 SLP-2단백대결장암 HCT-116세포증식화세포주기적영향。방법채용면역조직화학법검측50례결장암급대응암방정상결장조직중 SLP-2단백적표체，분석결장암병례림상병리특정여SLP-2단백표체적관계，장 SLP-2소간우 RNA（siRNA）전염 HCT-116세포，채용역전록（RT）-PCR급 Western blot검측전염SLP-2 siRNA후 HCT-116세포중기인급단백적표체정황，MTT법화류식세포술분별검측전염 SLP-2 siRNA후 HCT-116세포생장상황급세포주기。결과암방정상결장조직급결장암조직중 SLP-2적고표체솔분별위22％화70％，차이유통계학의의（P＜0．05）；SLP-2표체정황여결장암분기급림파결전이정황상관（P＜0．05）。전염 SLP-2 siRNA후，HCT-116세포중 SLP-2 mRNA화단백적표체균하강（P＜0．05），HCT-116세포 G1기증고，S기감소，세포생장변만（P＜0．05）。결론 SLP-2재결장암중고표체，병삼여결장암적발생발전。
Objective To investigate SLP-2 expression in colorectal cancer and its influence on proliferation and cell cycle of HCT-116 cells.Methods Immunohistochemical method was used to detect the expression of SLP-2 in 50 normal colorectal tissues and colorectal cancer tissue specimens.Then,analysis the relationship between the expression of SLP-2 and the clinical pathological characteristics of the 50 colorectal cancer cases was made.SLP-2 siRNA was transfected into colorectal cancer cell line HCT-116. RT-PCR and Western blot were used to detect the SLP-2 gene expression in HCT-1 1 6 after the transfection.The cell proliferation was detected by MTT,the cell cycles was analyzed by flow cytometry.Results SLP-2 expression in colonic tissue and colorectal cancer tissue were 20% and 70% respectively,which showed statistical significance(P<0.05).The expression of SLP-2 in colorec-tal cancer tissues was associated with TNM stage and lymph nodes metastasis(both P<0.05).After SLP-2 siRNA was interfered, the HCT-116 cells showed slower growth,the number of cells in G1 phase was increased and S phase was declined(both P<0.05), And the expression of SLP-2 in HCT-116 cell was decreased(P<0.05).Conclusion SLP-2 protein shows up-regulation in colorec-tal cancer,and it may have some relationship with carcinogenesis and development of colorectal cancer.
